| Objective: The aim is to provide more theoretical evidence for the application of ciRS-7in the diagnosis,treatment and prognosis of gastric carcinoma via studying the expression and correlation of ciRS-7,miR-7 and FAK in gastric carcinoma tissues and matched non-tumor tissues.Methods: The expression of ciRS-7,miR-7 and FAK m RNA in gastric carcinoma tissues and matched non-tumor tissues is detected by q RT-PCR,and the protein of FAK is detected by immunohistochemical staining.The correlation between their expression and the relationship between them and the clinicopathological features of gastric carcinoma are analyzed by using statistical software.Results:(1)The expression of ciRS-7 in gastric carcinoma tissues was higher than that in matched non-tumor tissues,and the expression level was related with the degree of invasion,lymph node metastasis and clinical stage(P<0.05),but not with age,gender and the degree of differentiation(P>0.05).The expression of miR-7 in gastric carcinoma tissues was lower than that in matched non-tumor tissues(P<0.05),and the expression of FAK m RNA in gastric carcinoma tissues was higher than that in matched non-tumor tissues(P<0.05),and they were all associated with tumor differentiation,invasion,lymph node metastasis,and clinical stage(P<0.05),but not with age and gender(P> 0.05).(2)The positive expression rate of FAK protein in gastric carcinoma tissues was higher than that in matched non-tumor tissues(P<0.05).The relative expression of FAK m RNA in gastric carcinoma tissues that associated with positive expression of FAK protein was significantly higher than that in negative expression(P<0.05),and the difference between them was statistically significant.(3)there was a negative correlation between ciRS-7 and miR-7expression in gastric carcinoma tissues(r =-0.9137,P < 0.0001).The expression of ciRS-7and FAK m RNA was positively correlated in gastric carcinoma(r = 0.8627,P < 0.0001),the difference was statistically significant.Conclusion: The expression of ciRS-7 and FAK m RNA and protein in gastric carcinoma tissues is up-regulated compared with matched non-tumor tissues.The expression of miR-7in gastric carcinoma tissues is down-regulated compared with matched non-tumor tissues,and the expression of ciRS-7,miR-7 and FAK is correlated with the degree of invasion,lymph node metastasis and clinical stage.The combined detection of the three is helpful to evaluate the invasion and metastasis of gastric cancer and to judge the prognosis.The expression level of ciRS-7 and miR-7 is negatively related,and the expression level of ciRS-7 and FAK m RNA is positively correlated.It’s indicated that ciRS-7 may increase FAK and promote the EMT through combining with miR-7 competitively.It’s may involved in the early stage invasion and metastasis of gastric carcinoma. |
