Rheumatoid arthritis(RA)is an autoimmune disease that seriously threatens human health.At present,seeking effective functional factors from food materials with good activity to ameliorate RA has received extensive attention.Recently,Dendrobium huoshanense has been approved as an edible plant,which has been used for the treatment of arthralgia syndrome in traditional Chinese medicines.In the light of polysaccharides as the main active components of D.huoshanense,the improvement effect of polysaccharide(cDHPS)isolated from Dendrobium huoshanense stems on RA in collagen-induced rheumatoid arthritis(CIA)mice was investigated in order to provide a theoretical basis for the application of D.huoshanense polysaccharides in ameliorating RA.The research contents and the obtained results are as follows:(1)cDHPS could improve the apparent indexes of CIA mice.The result of body weight indicated that cDHPS could improve weight loss due to RA and contribute to growth of CIA mice;The results of organ index showed that cDHPS could relieve the swelling of thymus,spleen,lung and liver as well as kidney injury in CIA mice;The rota rod test showed that cDHPS could enhance the pain tolerance of CIA mice;The results of arthritis score and paw thickness showed cDHPS could significantly reduce the swelling and distortion of joints in CIA mice compared with the model group;The3D modeling diagram of micro-CT showed that cDHPS could reduce the rate of bone opening in CIA mice and protect bone tissue.(2)cDHPS could improve the pathological indexes of CIA mice.H&E staining and CD90 immunofluorescence staining showed that cDHPS inhibited fibroblast proliferation;The levels of VEGF and CD31 indicated cDHPS reduced angiogenesis in CIA mice;The results of MMP2,MMP3,MMP8 and MMP9 content as well as OPG and RANKL immunohistochemical showed cDHPS could reduce the erosion of cartilage and bone;The results of cytokines detection and immunofluorescence of Th17 and Treg cells showed that cDHPS could inhibit inflammatory response and played an immunomodulatory role in CIA mice.(3)cDHPS could inhibit inflammatory signaling pathways in CIA mice.The effect of cDHPS on the inflammatory signaling of NF-κB and PI3K/AKT was detected by western blot.The results suggested cDHPS relieved inflammation by inhibiting the phosphorylation of IκB,p65,PI3 K and AKT,which resulted in the inhibition of NF-κBand PI3K/AKT signaling pathways as well as the reduction of inflammatory factors secretion in CIA mice.In summary,cDHPS could improve the symptoms of rheumatoid arthritis in mice,which may be achieved by inhibiting inflammatory signaling pathways and reducing secretion of inflammatory factors.
