Effects Of Huazhuo Jiedu Shugan Decoction On Learning And Memory Behavior And Expression Of AMPA Receptor GluR2 In Rats Model Of Epilepsy

Objectives By observing the effect of Huazhuo Jiedu Shugan decoction on the expression of AMPA receptor GluR2,seizure grade,seizure latency,Morri water maze and Y-maze in CA1 and CA3 regions of hippocampus of pentylenetetrazol-induced epileptic rats,the mechanism of anti-epileptic action of Huazhuo Jiedu Shugan decoction was explored,which laid a foundation for its clinical application.Methods 60 rats of SD were selected,male.Twenty rats were selected as non-model group by random number table method,and the other forty rats were used pentetrazol(PTZ)chemical kindling method to establish epilepsy model.Finally,the rats were randomly divided into six groups: blank saline group,blank Chinese medicine(high dose)group,model group,low dose group,high dose group and sodium valproate group.The state of the model was determined by Lado classification method.After 21 days of treatment with different doses of Huazhuo Jiedu Shugan decoction,the seizure grade,seizure latency and two behavioral differences were observed.Maze experiment includes Y-maze experiment and Morris water maze experiment.The expression of AMPA receptor GluR2 in CA1 and CA3 of hippocampus was detected by immunofluorescence.Results 1 Seizure grade observation: there was no seizure in the blank saline group and blank Chinese medicine group during the experiment.The seizure grade of rats in the high dose group and the sodium valproate group was significantly lower than that in the model group(P < 0.01).2 The latency of epileptic seizure: there was no epileptic seizure in the blank saline group and blank Chinese medicine group.The latency of epilepsy in the high dose group and the sodium valproate group was significantly higher than that in the model group(P < 0.01).3 Y-maze experiment: after treatment,the data of error avoidance times of two days were measured.On the first day,there was no statistical significance in each group(P > 0.05);on the second day,the error avoidance times of sodium valproate group and high dose group of Huazhuo Jiedu Shugan decoction were significantly lower than that of model group(P < 0.01).4 Morris water maze experiment: there was no statistical significance between the first time and the second time(P > 0.05);comparison of the third time: the escape latency of rats in the sodium valproate group and the high dose group of Huazhuo Jiedu Shugan decoction was significantly lower than that in the model group(P < 0.01).The times of crossing platform: the times of crossing platform in sodium valproate group and high-dose Huazhuo Jiedu Shugan decoction group were significantly higher than that in model group(P < 0.01).5 Immunofluorescence results: compared the number of positive cells of AMPA receptor GluR2 in the brain of rats in each group after treatment,the number of positive cells in CA1 and CA3 areas of hippocampus: the high dose group of Huazhuo Jiedu Shugan decoction and the sodium valproate group was significantly higher than that in the model group(P < 0.01).Conclusions 1 Huazhuo Jiedu Shugan decoction can significantly reduce the seizure grade and prolong the seizure latency of PTZ-induced epilepsy rats.2 Huazhuo Jiedu Shugan decoction can significantly improve the learning and memory ability of PTZ-induced epilepsy rats.The improvement effect is stronger than simple western medicine treatment.3 Huazhuo Jiedu Shugan decoction can significantly increase GluR2 protein expression in the hippocampus of epilepsy rats,suggesting that Huazhuo Jiedu Shugan decoction may play a role in improving seizures and learning and memory by up-regulating the expression of GluR2.4 Through behavioral and immunofluorescence observation,it was found that the expression of blank Chinese medicine group was similar to that of blank saline group,suggesting that Huazhuo Jiedu Shugan decoction would not cause learning and memory or cognitive impairment in rats.Figure 4;Table 8;Reference 136...
