| Purpose:Nasal mucosal epithelial barrier,as the body’ s first line of defense,is closely related to the occurrence,progression and improvement of allergic reactions.The destruction of the epithelial barrier of the nasal mucosa may be an important part of the pathogenesis of allergic rhinitis.Restoring the integrity of the barrier structure and function may be an effective method for the treatment of allergic rhinitis.From this we guessed that the effect of catgut embedding at acupoints on allergic rhinitis may be related to the positive regulation of the nasal mucosal epithelial barrier.The tight junction is located at the top of the adjacent surfaces of two epithetial cells and seals the gap between the basolateral membrane,which plays a decisive role in the mucosal barrier function.This study started with the tight junctions of nasal mucosal epithelial cells,speculated that the nasal mucosal barrier function changed,and explored the mechanism of acupoint thread embedding on allergic rhinitis.And according to the expression of Ctaudin-1 and Occludin to seek possible mechanisms of tight junction changes.Method:Purchase 40 healthy SD rats and randomly allocate them to the blank group,AR model group,fluticasone propionate group,acupoint embedding group,and sham embedding group,with 8 rats in each group.All rats except the blank group were prepared AR rat models according to the ovalbumin modeling method.After the model is complete,the AR model group and the blank group will not be intervened;the fluticasone propionate group will be intervened with fluticasone propionate once a day for 2 consecutive weeks;the acupoint catgut embedding group will be catgut-embedded at Yingxiang(LI20)once a week,For 2 consecutive weeks;false thread embedding in the false thread embedding group,no thread embedding,and the same time as the thread embedding group.Score and record AR-related behaviors in rats.ELISA was used to detect rat serum sIgE.After HE staining,the inflammatory response of nasal mucosa was observed under light microscope,and the tight junctions of nasal mucosa epithelial cells and the ultrastructure of endoplasmic reticulum were observed under transmission electron microscope.And measure the width of tight junction,WB method to measure the expression of tight junction protein Claudin-1 and Occludin.Result:1.Rat allergic rhinitis-related behavioral score contrast:when opposed to the blank group,the AR model group and the false-embedded group allergic rhinitis-related symptom scores were significantly increased,with statistical difference(P<0.05);and compared with the AR model group and the false catgut embedding group,the scores of catgut embedding group and the fluticasone propionate group were significantly lower,the imparity was statistically significant(P<0.05),and without notable difference between the groups(P>0.05).2.The results of ELISA detection of serum sIgE showed that the results of the AR model group and the false-embedded group showed that the serum sIgE level was significantly increased,and without notable difference between this two groups(P>0.05).However,both the catgut embedding group and the fluticasone propionate group can signifi-cantly reduce the serum sIgE value(P<0.05),and without notable difference between the groups(P>0.05).3.HE staining of nasal mucosa tissue under light microscope show ed that the nasal mucosal cilia in the model group and the false embedding group were arranged chaotically and fell down,and there were a large number of lymphocytes and eosinophils.When opposed to the model group,the inflammatory reaction was lighter in the catgut embedding group and the fluticasone propionate group.4.Tight connection between epithelial cells under transmission electron microscope:the tight connection of the blank group was a continuous narrow band;the tight connection of the model group and the false buried wire group was severely damaged,irregularly expanded,continuous interruption,and the width of the tight connection increased(P<0.05);the buried wire group of the acupoint and fluticasone propionate The tight connection of the group tends to be regular,and the width decreases compared with the model group and the false-embedded group(P<0.05).5.The endoplasmic reticulum morphology of epithelial cells under the transmission electron microscope:the endoplasmic reticulum in the model group and the pseudo-embedding group were swollen and expanded,showing vacuole-like changes,and the ribosomes were obviously shed.Pathological conditions improved.6.WB results showed that in the comparison of the expression levels of Claudin-1 and Occludin in each group,the expression of the false-embedded group and the model group was significantly reduced,and without notable statistical difference between the two groups(P>0.05).Both catgut embedding group and fluticasone propionate group can promote the expression of Claudin-1 and Occludin protein,and without notable statistical difference between the two groups(P>0.05).Conclusion:1.The expression of tight junction proteins Claudin-1 and Occludin is decreased,the tight junction is breaked down,and the function of the nasal mucosal epithelial barrier is weakened,which is conducive to the transmission of allergens across the epithelium and triggers an allergic reaction,perhaps it is the key mechanism of AR pathogenesis.2.Catgut embedding at acupoints can reduce the serum sIgE of AR rats,relieve the inflammatory reaction of the nasal mucosa,significantly reduce the AR-related symptoms,and without significant statistical difference compared with fluticasone propionate treatment(P>0.05).3.Acupoint embedding can increase the levels of tight junction proteins Claudin-1 and Occludin,improve the tight junction of nasal mucosa epithelial cells,enhance the guard ability of nasal mucosa,thereby reducing the invasion of allergens,alleviating the symptomsof allergic rhinitis,puting a positive effect to the treatment of allergic rhinitis. |
