Insulin resistance,as a kind of obesity,type 2 diabetes and metabolic syndrome is the key cause of nonalcoholic fatty liver disease,its incidence trend of increased year by year and tend to be younger ages,serious harm human health and cause great economic burden,in recent years has become a big difficulty in the world health organization(who),one of the so for the prevention and treatment of insulin resistance is very important.The early stage of the research study found that sulforaphane(SFN)potentially regulate glucolipid metabolism effect,in this paper,by the high fat diet rats with insulin resistance model is set up,SFN foodborne phytochemicals is added in the diet,with intestinal flora as the breakthrough point,to explore its to the intestinal mucosal barrier and the effect of short chain fatty acids related pathways and mechanisms,from the perspective of dietary interventions for the prevention and treatment of insulin resistance glucolipid metabolic diseases such as to provide scientific basis.The rat model of insulin resistance was established by high-fat diet.The experiment was divided into four groups: normal group(CK),high-fat diet group(HFD),SFN medium-dose group(SM),and SFN high-dose group(SH).The whole experiment lasted for 10 weeks.The relative indexes of serology were determined by ELISA.High throughput sequencing of 16 Sr DNA was used to characterize intestinal flora.Determination of short chain fatty acids by gas chromatography;Rt-pcr was used to determine the gene expression level of tight junction protein and the number of lactobacillus,bacteroidetes and bifidobacteria.The expression of short chain fatty acid receptor protein was determined by Western blot.SFN can reduce the weight of insulin-resistant rats,reduce serum TG,TC and insulin content,improve the ability of rats to empty blood glucose and insulin tolerance,increase insulin sensitivity,and reduce the status of insulin resistance;SFN can increase the proportion of probiotics in the intestinal tract of insulin rats,reduce the Bacteroidetes/Firmicutes value(B/F value),and increase the abundance of Bacteroidetes,Bifidobacteria and Butyricimonas etc.of short-chain fatty acid producing bacteria.It had a remodeling effect on the intestinal flora of insulin-resistant rats and improved the intestinal flora disorder caused by high-fat diet.SFN can reduce the content of serum LPS in rats,increase the m RNA expression of tight junction protein zo-1 and occludin as well as the number of lactobacillus,bifidobacterium and bacteroides,maintain the stability of intestinal mucosal barrier and improve intestinal permeability.SFN can activate the scfas-gpr-gastrointestinal hormone pathway in insulinresistant rats,increase the contents of acetic acid and butyric acid in intestinal tract,up-regulate the expression of SCFAs receptor proteins GPR41 and GPR43,and increase the secretion of gastrointestinal hormone GLP-1.In conclusion,SFN can improve insulin resistance of rats induced by high-fat diet by regulating intestinal flora,protecting intestinal mucosal barrier,and regulating scfas-gpr-gastrointestinal hormone-related pathways.
