In recent years,due to its miniaturization,simple operation,immediate reporting results,and low cost,POCT(point-of-care testing)has attracted widespread attention from clinical departments,community clinics,primary hospitals,and patient families.Based on the current research status of specific immune protein POCT products at home and abroad,this paper designs and develops a specific protein detection system that is easy to operate,fast and small in size:Based on the principle of Lambert-Beer law,the transmission colorimetry method(transmission turbidimetry)is adopted,using the optical part,electronic part and mechanical part as the structure,carring out the photoelectric design part and software design part.The optical design part divides the light path of the halogen lamp light source into four parallel light paths after convergence and collimation,using optical fiber to transmit the light,which can test 1~4 items;In the electronicdesign part,STM32 microcontroller is used as the main control chip,adopts temperature control module for constant temperature control,use photovoltaic cells,A/D acquisition modulesand photoelectric processing modules to obtain the voltage value changes during the reaction and converted into absorbance values;Using the built semi-automatic specific immune protein POCT rapid detection system,to establish a standard curve for commercial specific protein standards fitted by designed software processes by this system;Appling the system to the rapid urine detection of four specific immune proteins POCT for kidney injury,and the experimental comparison with automatic biochemical analyzers at home and abroad was performed.The system verification data shows: The absorbance change of every standard solution of this system is not more than ± 0.005,and the detection CV values are less than 1%,indicating that the stability,repeatability and linearity of the instrument have met the requirements.All functions in this design can be realized.Test results of urine specific immune protein concentration of four items of kidney injury(cystatin C,microalbumin,NGAL and creatinine)shows,urine is used as the sample,which is convenient,the test results can be obtained in 5~10 minutes.The standard curve R2 value of this system can reach above 0.993;The detection range of absorbance is 0.01~1.8,and the CV value of the standard solution is less than 1%.Using this system to practical application test of human urine was performed,the maximum value of microalbumin CV was 2.61% and the maximum value of creatinine CV was 3.17%,in the two tested items,both of which were less than 3.5%,the experimental results reached expectations.Compared with the Uicare-300 POCT automatic urine immunoanalyzer and Hitachi automatic biochemical analyzer on the market,the test values of the tested microalbumin and creatinine were basically the same.The specific immune protein POCT system designed and developed in this paper canbe applied to the detection of kidney injury indicators,as well as rapid detection of myocardial indicators,blood lipid indicators,etc.The results of this study provide an experimental basis for the development and clinical application of specific immune protein POCT technology. |
