Cercical Cancer is the fourth leading cause of gynecological malignancy in the world.The International Angency for Reasearch on Cancer(IARC)reports that the number of new cases of cervical cancer in the world has reached 57000 and the number of deaths has reached 310000.WHO(World Health Organization)listed cervical cancer as one of the world’s top ten public health security threats.Rates vary significantly by region,with the vast majority of cervical cancer cases coming from the developing counties.In China,the incidence and mortality of cervical cancer are significantly higher than the world average.Relevant date showed that the incidence(6.6%)and mortality(7.5%)of cervical cancer in chinese women remained high in 2018,and women of childbearing age accout for the majority of them.With the deepening of scientific reaserch,High-risk human papillomavirus(HPV)and cervical cancer related factors of the disease also gradually revealed.HPV vaccine has brought countless women with the advent of hope,because of various reasons,the incidence of cervical cancer still according to the second place for female reproductive system tumor.At present,the main treatment methods for cervical cancer patients include surgery,radiotherapy and chemotherapy.Individualized treatment plans are formulated for different patients.Most of the patients with early cervical cancer should be treated with surgery,supplemented by radiotherapy and chemotherapy,while those with advanced cervical cancer should be treated with chemoradiotherapy.The adverse consequences brought by radiotherapy and chemotherapy have seriously affected the quality of life of the patients.The reasearch on the molecular mechanism of cervical cancer is becoming more and more in-depth,including the insertion of different cervical cancer RNA,epithelial-mesenchymal transformation,signaling pathways of different action sites and so on.Therefore,in order to further study the pathogenesis of cervical cancer,we provide new ideas and methods for the diagnosis and treatment.Objective:To observe the expression of PRR11 and SKA2 protein in cervical cancer tissues,and to analyze the relationship between PRR11 and SKA2 protein and the clinical pathological parameters and prognosis of cervical cancer.Statistical methods:SPSS statistical sofeware was used for data analysis,χ~2 test was used to analyze its expression in cervical cancer tissues and adjacent normal tissues.The correlation between PRR11 and SKA2 expressions was analyzed by Spearman method,the survival rate among groups with different protein expressions was analyzed by kaplan-meier,and the prognostic factors were analyzed by Cox regression proportional risk model.Experimental materials and methods:Tissue specimens of 289 patients with cervical cancer who underwent surgical resection in Yuhuangding hospital,Yantai affiliated to Qingdao university from January 2012 to December 2015 and their adjacent normal tissues were collcected.Immunohistochemistry was used to detect the positive expression rates of PRR11 and SKA2 in 289 cervical cancer tissues and the adjacent normal tissues,to investigate the relationship between them and the clinicopathological characteristics of cervical cancer patients,Cox single factor and multiple analysis PRR11 and SKA2 and the correlation between the prognosis of patients with cervical cancer.Results:PRR11 in cervical cancer tissue and normal tissue adjacent to carcinoma positive expression rate was 60.21%(174/289)and 16.96%(49/289),SKA2 in cervical cancer tissues and in normal tissue adjacent to carcinoma positive expression rate was 69.55%(201/289)and 37.37%(108/289),cervical cancer tissues PRR11 and SKA2 positive expression rate is significantly higher than tissue adjacent to carcinoma(60.21% VS 16.96%,χ~2=1114.081,P<0.001;69.55% VS 37.37%,χ~2=60.143,P<0.001),statistically significant difference(P<0.05).Spearman correlation analysis results showed that PRR11 was positively correlated with SKA2 expression(r = 0.725,P<0.001).The exprssion of PRR11 and SKA2 proteins was related to FIGO stage,tissue differentiation degree and lymph node metastasis(P<0.05),but there was no related to age,tumor diameter,muscle infiltration and vascular infiltration(P>0.05).The 3-year OS of patients with positive exprssion of PRR11 and SKA2 were significantly lower than those with negative exprssion(78.16%VS94.78%,P<0.05;82.09% VS 90.91%,P<0.05).Cox regression analysis showed that the factors affecting the prognosis of patients with FIGO stage Ⅲ-Ⅳ period,PRR11 positive expression and SKA2 positive expression is independent risk factor for cervical cancer with poor prognosis(HR=1.698,95%CI:1.369-2.876,P<0.001;HR=0.418,95%CI:0.224-0.714,P<0.001;HR=0.626,9 5%CI:0.437-0.915,P<0.001).Conclusion: 1.PRR11 is highly expressed in cervical cancer.The higher stage,the higher positive expression rate and it is related to the degree of tissue differentiation and the pressence or absence of lymph node metastasis.2.SKA2 is highly expressed in cervical cancer.The higher stage,the higher positive expression rate and it is related to the degree of tissue differentiation and the pressence or absence of lymph node metastasis.3.PRR11 and SKA2 are highly expressed and positively correlated in cervical cancer,which are independent risk factors affecting the prognosis of patients and may become effective indicators for the diagnosis and prognosis assessment of cervical cancer.
