Objective: Osteosarcoma(OS)is a malignancy that originates in the mesenchymal tissue and has the highest incidence and mortality among primary bone tumors.Therefore,it is of great significance to explore the mechanism of osteosarcoma occurrence and progression and find more sensitive molecules as therapeutic targets.In this thesis,the expression,function and prognosis of PRR11 in osteosarcoma tissues and cells were analyzed by bioinformatics.The effects of PRR11 on proliferation,invasion,migration and apoptosis of osteosarcoma cells were explored through cell experiments.To provide a research basis for PRR11 as a therapeutic target for osteosarcoma.Methods: The GEPIA database was used to explore the expression level of PRR11 in malignant tumor tissues,and the CCLE database was used to explore the expression level of PRR11 in malignant tumor cells.UCSC Xena database obtained transcriptome data and clinical samples for osteosarcoma,and divided the samples into two groups,high and low expression groups,based on the median PRR11 expression,to explore the effect of PRR11 on the prognosis of osteosarcoma patients.Gene set enrichment analysis(GSEA)was used for exploring the biological functions and pathways of the PRR11 differentially expressed genes.Protein-protein interaction(PPI)network was used to explore the binding sites of PRR11 in osteosarcoma.After upregulating PRR11 in MG-63 cells of osteosarcoma,the role of PRR11 in osteosarcoma was further investigated by CCK-8 assay to detect cell proliferation,Transwell to detect cell invasion,scratch assay to detect cell migration,and flow cytometry to detect apoptosis.Results: PRR11 is highly expressed in various malignant tumors,and is also highly expressed in osteosarcoma tissues and cells.Patients with osteosarcoma with high PRR11 expression had significantly lower survival than those in the low expression group.Cox regression analysis found that PRR11 was a risk factor for prognosis.The GSEA results showed that the differential genes were mainly enriched in ubiquitin-mediated proteolysis and cell cycle biological processes.The PPI network mainly interacted with the regulatory genes of the cell cycle.PRR11 promotes the invasion,migration,and proliferation of osteosarcoma cells and inhibits their apoptosis.Conclusion: This study preliminarily confirmed that PRR11 is highly expressed in osteosarcoma and has a significant impact on prognosis,and PRR11 is involved in regulating the malignant biological behavior of proliferation,migration,invasion and apoptosis of MG-63 cells in osteosarcoma.PRR11 as a cell cycle regulatory protein that promotes osteosarcoma initiation and progression may be related to ubiquitin proteasome-mediated cell cycle regulation.Therefore,PRR11 as an oncogene promotes the occurrence and development of osteosarcoma,which may become a prognostic index and therapeutic target for evaluating osteosarcoma in patients.
