Objective: CEACAM6 regulates the occurrence and development of digestive system tumors through multiple pathways,but its role in gallbladder cancer has not been reported.This study aimed to investigate the effect of CEACAM6 gene silencing on the proliferation,migration,invasion and apoptosis of gallbladder cancer cell lines.Provide evidence for further basic gallbladder cancer research.Methods:GBC-SD and SGC-996 cell lines were tested in logarithmic growth.Two different siRNA recombinant expression vectors were established using CEACAM6 as a target for different sites.Quantitative real-time PCR(qRT-PCR)and Western blot analysis were used to detect the mRNA and protein expression of CEACAM6,respectively.CCK-8 assay and colony formation assay was performed to evaluate the cell viability.The variations of cell migration and invasion were detected by wound healing assay and Transwell assay,and detected the expression of invasion-related proteins in cells after silencing of CEACAMA6.Flow cytometry was applied to measure the cell apoptosis and cell cycle distribution,an detected apoptosis-related protein changes after silencing of CEACAM6.Results:The expression of CEACAM6 mRNA and protein in the CEACAM6 siRNA group was significantly lower than that in the NC group and the blank group.Compared with the NC group,the cell proliferation of CEACAM6 siRNA group was significantly reduced,but the apoptosis rate was significantly increased.The expression of apoptosis-related protein Bax in CEACAM6 siRNA group was significantly decreased,expression of anti-apoptotic protein Blc-2 was increased significantly.In addition,in the CEACAM6 siRNA group,the percentage of cells in the G0/G1 and G2/M phases gradually increased,but decreased in the S phase.The migration and invasion ability of gallbladder cancer cells was significantly lower than that of NC group.Compared with the NC group,the expression of MMP2 and Vimentin in CEACAM6 siRNA group was significantly reduced.Conclusion:Our study showed that CEACAM6 gene silencing could promote apoptosis of gallbladder cancer cells and inhibit proliferation,migration and invasion ofgallbladder cancer cells.
