Expression And Significance Of CEACAM6 In Gallbladder Carcinoma

Objective:The abnormal expression of CEACAM6 is considered as a potential biomarker in a variety of malignant tumors,but its role in gallbladder cancer is limited.The purpose of this study was to investigate the expression and significance of CEACAM6 in gallbladder carcinoma from the angle of correlation between histologic and clinicopathological factors.Methods:Cholecyst tissue microarray containing complete clinicopathological features and long-term follow-up data of 40 patients was selected.Immunochemistry was used to evaluate CEACAM6 expression in 40 GC specimens and 20 peritumoral tissue specimens.The correlation between CEACAM6 and histological or clinical factors was calculated byχ~2 independence test,and the correlation between survival and CEACAM6 positive expression and other clinicopathological features was analyzed by Kaplan-Meier survival analysis and multivariate cox regression analysis.Results:In 40 cases of gallbladder carcinoma,the expression of CEACAM6 was positive in 9 cases(100.0%)with lymph node metastasis,while it was positive in 20cases(64.5%)and negative in 11 cases(35.5%)in 31 cases without lymph node metastasis,the expression of CEACAM6 in gallbladder carcinoma was significantly correlated with lymph node metastasis(p<0.05).The expression of CEACAM6 was not significantly associated with sex,age,pathological type,pathological classification,invasion status,and distant metastasis(all P>0.05).The expression of CEACAM6 was positive in 4 cases(20.0%)and negative in 16 cases(80.0%)of 20peritumoral tissues.In 20 cases of GC tissues,13 cases were CEACAM6 positive(65%)and 7 cases were CEACAM6 negative(35%).The expression of CEACAM6was significantly greater in GC tissues than in peritumoral tissues(P<0.05).Of the 40follow-up patients,9 survived more than one year after operation,and 31 died within one year after operation.The mean survival time was 10.35±10.76 months.Of the 40patients,29 were positive for CEACAM6.The median survival time for patients with positive expression of CEACAM6 was 6 months,and the median survival time for patients with negative expression of CEACAM6 was 16 months.The median survival time in patients with lymph node metastasis was 2 months,and the median survival time for patients with no lymph node metastasis was 8 months.The median survival time for patients with distant metastasis was 3 months,and the median survival time for patients with no distant metastasis was 8 months.Kaplan-meier survival analysis showed that the total survival time was significantly correlated with the positive expression of CEACAM6,lymph node metastasis and distant metastasis in patients with gallbladder carcinoma(p<0.05),but was not significantly associated with sex,age,pathological type,pathological classification,and invasion status(all P>0.05).Multivariate cox regression analysis showed that survival time was significantly correlated with lymph node metastasis(p<0.05),but not with CEACAM6 positive expression and distant metastasis(p>0.05).Conclusions:Expression of CEACAM6 was significantly greater in GC tissues than in peritumoral tissues,and its positive expression was significantly correlated with lymph node metastasis.The positive expression of CEACAM6 may not affect the survival time of gallbladder carcinoma patients,or it may not be able to detect the survival difference between different CEACAM6 expression results due to insufficient sample size.
