When aluminum accumulates excessively in the bones,it can cause Osteoporosis.Traditional Chinese medicines(TCM)have shown good efficacy in the treatment of Osteoporosis,with little side effects,but the treatment mechanism have not been fully explained.Studies have shown that osteoporosis is closely related to aluminum overload in body,and there is no related research on the mechanism of aluminum flooding by commonly used TCM.From the perspective of aluminum overload,it is believed that certain components in TCM can complex with aluminum ions in the body to form complexes,which make the excess aluminum in the body reduce.Zebrafish is widely used in bone disease due to its advantages of high similarity to human gene bone development,ability to maintain the integrity of in vivo animals.In this paper,a rapid and efficient Osteoporosis model of zebrafish was established by using aluminum chloride as a model drug for the first time.The model was used to study the effect of Alpinia oxyphylla Miq.,Panax ginseng C.A.Mey.,Codonopsis pilosula(Franch.)Nannf.,Curcuma wenyujin Y.H.Chen et C.Ling,Amomum kravanh Pierre ex Gagnep.,Houttuynia cordata Thunb.,which have strong aluminum complexing ability,on preventing the loss of zebrafish bone.The results of previous experiments were further verified to provide research basis,and to provide experimental methods and basis for further elucidating the mechanism and material basis of the treatment of Osteoporosis by TCM.In the experiment,zebrafish(3 dpf)were cultured with different concentrations of aluminum chloride(5,10,15,20 ?g/ml)after fertilization.The alizarin red and alcian blue staining were performed at 7 dpf,and the bone mineralization area,accumulated optical density(IOD)and cartilage staining area were counted.On the basis of the above 5 ?g/ml aluminum chloride culture to 7 dpf,the aluminum chloride was removed,and then the positive control drug deferoxamine and different concentrations of Alpinia oxyphylla Miq.,Panax ginseng C.A.Mey.,Codonopsis pilosula(Franch.)Nannf.,Curcuma wenyujin Y.H.Chen et C.Ling,Amomum kravanh Pierre ex Gagnep.,Houttuynia cordata Thunb.,Decoction(0.1,1,10,50,100 ?g/ml)were used to culture to 11 dpf was stained with alizarin red and alcian blue,and bone mineralization area,accumulated optical density(IOD)and cartilage formation area were counted.The results showed that after cultivating zebrafish with different concentrations of aluminum chloride,the zebrafish bone mineralization area,cumulative optical density(IOD)and cartilage formation area decreased in a dose-dependent manner,and the difference was statistically significant compared with the control(p<0.001).50 ?M of desferrioxamine can prevent zebrafish bone loss and repair cartilage damage after high aluminum culture.Alpinia oxyphylla Miq.(0.05,0.1,0.5 ?g/m L),Panax ginseng C.A.Mey.(0.1 ?g/m L),Curcuma wenyujin Y.H.Chen et C.Ling(0.1,1,10,50,100 ?g/m L),Codonopsis pilosula(Franch.)Nannf.(0.1,1,10 ?g/m L),Amomum kravanh Pierre ex Gagnep.(10,50,100 ?g/m L)can prevent aluminum chloride-induced loss of zebrafish bone mass;Houttuynia cordata Thunb.(1,10 ?g/m L)can prevent aluminum chloride induced reduction of zebrafish bone mineralization.In this paper,the high aluminum-induced zebrafish osteoporosis model established which has obvious bone loss and cartilage damage,and can be used to screen osteoporosis drugs and their trace components;Deferoxamine can prevent bone loss in the established model,proved the feasibility of this model,and can choose it as a positive control drug of this model.Alpinia oxyphylla Miq.,Panax ginseng C.A.Mey.,Codonopsis pilosula(Franch.)Nannf.,Curcuma wenyujin Y.H.Chen et C.Ling,Amomum kravanh Pierre ex Gagnep.,Houttuynia cordata Thunb.can prevent bone loss of zebrafish induced by aluminum chloride.,which further proves the experimental results of early in vitro screening;And the complex with aluminum may be one of the mechanisms of Curcuma wenyujin Y.H.Chen et C.Ling,Panax ginseng C.A.Mey.,Houttuynia cordata Thunb.against osteoporosis;Alpinia oxyphylla Miq.,Codonopsis pilosula(Franch.)Nannf.,Amomum kravanh Pierre ex Gagnep.,may be potential drugs for the treatment of aluminum-induced osteoporosis.
