Allergic rhinitis(Allergic rhinitis,AR)is a non-infectious inflammatory disease of the nasal mucosa.It is an individual who is exposed to allergens and is involved with a variety of immune active cells and cytokines.Allergic rhinitis is closely related to other infectious diseases in the respiratory tract mucosa such as asthma,sinusitis,allergic conjunctivitis.About 400 million people worldwide are still suffering from the disease,and the prevalence rate continues to increase,the mechanism and epidemiology of allergic rhinitis and related diseases have always been the research hotspots.The pathogenesis of allergic rhinitis is mainly the activation of helper T cells 2(Thelper2cell,Th2)cells,Th2 cells overactivated and secreted cytokines such as IL-4,IL-5 and IL-15,which promoted the release of Ig E and induced subsequent hypersensitivity,and regulatory T cells(Treg)play an important role in the inhibition of Th2 cells.mi R-146 a is an important immunoregulatory factor with anti-inflammatory effect,and its expression in allergic rhinitis is reduced.Signal transduction and activator of transcription(STATs)act as molecules downstream of cytokines participate in the pathogenesis of autoimmunity and inflammatory diseases.And the role of STAT5 b in the maintenance and function of human Treg has been confirmed.The purpose of this study was to investigate the possibility of mi R-146 a affecting Treg cell proliferation by regulating the expression of STAT5 b and participating in the development of allergic rhinitis.This study used RT-PCR and western blot to detect the expression of mi R-146 a and STAT5 b in nasal mucosa tissue and peripheral blood mononuclear cells of the patients with allergic rhinitis,and used flow cytometry to detect the the proportion of Treg cells after inhibiting mi R-146 a expression in peripheral blood mononuclear cells,this study in order to provide new ideas for the pathogenesis of allergic rhinitis.Objective: 1.To investigate the expression of mi R-146 a and STAT5 b in the PBMC of patients with allergic rhinitis;2.To investigate mi R-146 a can influence the proliferation of Treg cells through STAT5 b in the peripheral blood of allergic rhinitis.Methods: 1.To collect nasal mucosa tissue from 15 cases of allergic rhinitis nasal mucosa tissue and 18 cases of control individuals,the expression of mi R-146 a was detected by PT-PCR method;2.To isolate peripheral blood mononuclear cells from 8 cases of allergic rhinitis nasal mucosa tissue and 8 control peripheral blood,PT-PCR was used to detect the expression of mi R-146 a and STAT5 b m RNA in PBMCs;3.To collect nasal mucosa tissue from 8 cases of allergic rhinitis and 8 cases of control individuals,the expression of STAT5 b was tested by western blot;4.Isolate peripheral blood mononuclear cells from 5 control individuals,the PBMC stimulated by IL-2 / IL-15,and transfected with Lentimi Ra-Off-hsa-mi R-146a-5p Virus,and the proportion of co-expression of lymphocytes and STAT5 b,the proportion of Treg cells and the proportion of the co-expression of lymphocytes and STAT5 b was detected by flow cytometry.Results: 1.The expression of mi R-146 a and STAT5 b in nasal mucosa tissue and peripheral blood mononuclear cells of patients with allergic rhinitis was lower than that in the control group;2.Inhibition of mi RNA-146 a expression in peripheral blood mononuclear cells of allergic rhinitis,the proportion of lymphocytes co-expressing STAT5 b was decreased,the proportion of Treg cells and the proportion of the co-expression of Treg cells and STAT5 b was decreased;Conclusion: 1.mi R-146 a and STAT5 b have low expression in nasal mucosa and peripheral blood of the allergic rhinitis;2.In peripheral blood,mi R-146 a has a positive effect on STAT5 b,mi R-146 a can affect the proliferation of Treg cells through STAT5 b.
