Objective: Lung cancer is a malignant tumor derived from the tracheobronchial mucosa or glands,the mortality and morbidity ranks first in China for malignant tumors.Non-small cell lung cancer(NSCLC)accounts for about 85% of the total incidence of lung cancer,adenocarcinoma accounts for about 40%,large cell carcinoma accounts for about 5-15%,and the remaining 30% is squamous carcinoma.For the molecular is very complex,and most of the patients are in the advanced stage when they are diagnosed,which makes the treatment very difficult.POLE2 is the second largest subunit of DNAP ε and plays an important role in the occurrence and development of rectal cancer,cervical cancer,bladder cancer,breast cancer and other malignant tumors.This experiment explores the expression of POLE2 in cancer tissues and corresponding adjacent tissues and its clinical significance in NSCLC.Method: 60 specimens of cancer tissues and corresponding adjacent tissues were collected from tangdou hospital of air force medical university from December 2016 to December 2018.All patients had no radiotherapy or molecular targeted therapy before surgery.We performed tumor staging according to the Chinese Medical Association’s Lung Cancer Clinical Diagnosis and Treatment Guidelines(2018 Edition),and all specimens were confirmed by pathological diagnosis.The distribution and expression of POLE2 in cancer tissues and adjacent tissues were detected by immunohistochemistry.At the same time,the serum tumor marker levels of the corresponding patients were detected by electrochemical luminescence method.The relationship between POLE2 expression level and cancer tissues and adjacent tissues,gender,age,smoking index,pathological type,difference degree of cancerous cells,lymph node metastasis,T stage,chemotherapy before surgery and tumor markers(CEA,CA-125,CYFRA21-1,Scc-Ag)were analyzed.Results:1.The positive expression rate of POLE2 is 58.33%(35/60)in non-small lung cancer tissues and 36.67%(22/60)in adjacent cancer tissues.The difference has statistical significance(P <0.05).2.The strong positive expression rate of POLE2 is 31.67%(19/60)in non-small lung cancer tissues and 16.67%(10/60)in adjacent cancer tissues.The difference has statistical significance(P <0.05).3.There is significant difference in the positive expression rate of POLE2 among non-small lung cancer tissue of smoking index and chemotherapy before surgery(P <0.05).4.There is no significant difference in the positive expression rate of POLE2 among non-small lung cancer tissue of different gender,age,pathological type,difference degree of cancerous cells,lymph node metastasis and T stage(P> 0.05).5.The serum CEA level in the positive expression is significantly higher than that in the negative tissues(P <0.05).Conclusion:1.The expression of POLE2 is high in the tissue of non-small lung cancer,but low in adjacent tissue of non-small lung cancer.2.The expression of POLE2 in non-small lung cancer tissues is related to the smoking index and chemotherapy before surgery but not to the gender,age,pathological type,difference degree of cancerous cells,lymph node metastasis and T stage.3.The concentration of serum carcinoembryonic antigen(CEA)in the positive expression group of POLE2 in NSCLC cancer tissues is high. |
