| OBJECTIVE Dipeptidyl peptidase-4 inhibitor(DPP-4i)is commonly used worldwide for the treatment of type 2 diabetes mellitus.In addition to its hypoglycemic activity,DPP-4i might have anti-inflammatory effects.In obese patients,macrophage-induced adipose tissue inflammation plays an important role.To establish a model of obese rats with type 2 diabetes by feeding a high-fat,high-sugar diet combined with a small dose of streptozotocin,and to randomly divide the model into experimental and control groups.The experimental group was divided into diabetic treatment group(DM-s group)and diabetic control group(DM group).DM-s group was given Saxagliptin intervention.Finally,by detecting the change of serum sCD163 in each group,and explored the effect of saxagliptin on anti-inflammatory in type 2 diabetic rats.METHOD Sixty male Sprague Dawley rats were randomly divided into the normal control group and the model group.Obese type 2 diabetic rats were induced by high-fat diet combined with streptozotocin and randomly divided into diabetic control group and saxagliptin treatment group.The treatment group was treated with saxagliptin for intervention.The fasting blood glucose was measured by blood glucose meter.The automatic biochemical analyzer was used to detect changes in blood lipids and fasting insulin.Elisa was used to detect serum sCD163,TNF-α,IL-1,IL-10 and adiponectin levels.RESULT(1)The indexes of blood glucose,blood lipids,and insulin resistance were significantly higher in the DM group and the DM-sax group than in the NC group.The above indexes in group DM were all lower than that in group DM,and the difference was statistically significant.(2)The Serum sCD163 levels were significantly higher in the DM group and the DM-sax group than those in the NC group.Serum sCD163 levels in the DM-sax group were lower than those in the DM group and the difference was statistically significant.There was no correlation between changes in sCD163 and changes in HbAlc and body weight.(3)The levels of TNF-α and IL-1β in DM group was higher than that in normal group.The TNF-α and IL-1β level in DM-sax group was lower than that in DM group,the difference was statistically significant.The levels of IL-10 and adiponectin in DM and DM-s group was lower than that in normal group.The IL-10 and adiponectin level in DM-sax group was higher than that in DM group,the difference was statistically significant.CONCLUSION With the development of animal models of type 2 diabetes with obesity,serum cholesterol,LDL,and sCD 163、TNF-α、IL-1β increased,and IL-10、adiponectin levels decreased.After intervention with saxagliptin,the IL-10 and adiponectin levels increased while the other indicators showed a downward trend,indicating that Saxagliptin can regulate the innate immune,induce macrophage differentiation,alleviate adipose tissue inflammation in the treatment of obese type 2 diabetes mellitus. |
