Objective To investigate the association of polymorphisms of FCGR II and III genes with susceptibility and clinical phenotype of systemic lupus erythematosus(SLE).Methods From January 2017 to December 2017,144 SLE patients were enrolled as the experimental group and 150 healthy persons as the control group.Blood samples,clinical data and laboratory examination data were collected.Genomic DNA was extracted by single base extension PCR and 27 loci were analyzed by mass spectrometry.SNP detection and SPSS software were used to analyze the relationship between Fc gamma receptor gene polymorphism and SLE susceptibility and clinical phenotype.Results ①The homozygote TT genotype frequencies at rs1050501 locus were 50.69%and 64.00%in the experimental group and the control group,respectively,with statistical significance(p=0.021).The frequency of heterozygote CT genotype at rs115878669 locus was 50.69%and 64.00%in the experimental group and the control group,respectively.The difference was statistically significant(p=0.021).The frequency of homozygote TT genotype was 23.61%and 10.00%in the experimental group and the control group,respectively.The difference was statistically significant(p=0.002).The homozygote TT genotype frequencies at rs147574249 locus were 17.36%and 8.00%in the experimental group and the control group,respectively.The difference was significant(p=0.016).The homozygote GG genotype frequencies at rs199705513 locus were 20.83%and 10.00%in the experimental group and the control group,respectively.The difference was significant(p=0.010).The heterozygote GA genotype frequencies were 59.03%and 70.00%in the experimental group and the control group,respectively The genotype frequencies of ho mo zygote GG at rs2333843 locus were 31.94%and 16.00%respectively in the experimental group and the control group(p=0.001).The genotype frequencies of heterozygote GC were 65.97%and 80.00%in the experimental group and the control group,respectively.The difference was significant(p=0.007),and the CA genotype frequencies of heterozygote at rs77717968 locus were real.There were significant differences between the experimental group and the control group(30.56%and 46.00%,respectively)(p=0.007).There was no significant difference between the other two groups.②In the experimental group,SLE patients such as CC genotype at rs1050501 locus and CT genotype at rs115878669 locus were more likely to have blood system involvement;GG genotype at rs114531649 locus,TT at rs12131862 locus,TC genotype,AA at rs144426992 locus,GA genotype,CC genotype at rs147574249 locus,AA genotype at rs199705513 locus,AA genotype at rs61803007 locus CC,TC genotype,and AA gene at rs77717968 locus were more likely to have blood system involvement.The AA genotype at rs144426992 locus and TC genotype at rs146653557 locus were more prone to serositis,but there was no significant difference in other comparisons.Conclusion 1.The genetic polymorphisms at rs1050501,rs115878669,rsl47574249,rs199705513,rs2333843 and rs77717968 loci of FCGRII and III genes are related to the susceptibility of SLE.2.In SLE group,the polymorphism of some loci was related to the clinical phenotype of SLE. |
