Objective:To observe the changes of the protein expression levels of collagen type I and collagen type Ⅲ and the ratio of collagen types Ⅰ/Ⅲ during wound healing and to explore the potential mechanism.Methods:A circular excisional,full-thickness acute wound,with a diameter of 20 mm was created on the back of thirty-six healthy male rat,all model rats were then randomized into two groups:the control(blank group,n=18)and receiving vacuum sealing drainage therapy(VSD therapy group,n=18)and each group was assigned into three subgroups based on their sacrificed time(named 1,3 and 7 day group,n=6 each group).we photographed wound to calculate and compare wound closure rate.The mRNA and protein expression levels of type Ⅰ collagen and type Ⅲ collagen and its ratio were detected by qRT-PCR,immunohistochemistry and Sirius scarlet staining.The structure of granulation tissue and the length of re-epithelialization were surveyed by histologyResults:1.Compared with the control group,the wound surface in the VSD group was moister.The wound closure rate in the VSD group was higher than that in the control group at days 1,3 and 7(P<0.05).2.The mRNA expression levels of type Ⅰ collagen and type Ⅲ collagen in the VSD group was higher than that in the control group at days 1,3 and 7(P<0.05).3.The results of immunohistochemistry(the average optical density value represented the protein expression levels of collagen type I and collagen type III)showed that:The protein expression levels of type I collagen in the VSD group was higher than that in the control group at days 1,3 and 7(P<0.05).The protein expression levels of type III collagen in the VSD group was higher than that in the control group at the first day(P<0.05).The ratio of collagen types Ⅰ/Ⅲ in the VSD group was higher than that in the control group at days 3 and 7(P<0.05).4.Sirius scarlet staining show that:The ratio of collagen types Ⅰ/Ⅲ in the VSD group was higher than that in the control group at days 3 and 7(P<0.05).5.H&E staining show that:The area of granulation tissue in the VSD group was higher than that in the control group at first day(P<0.05).The area of granulation tissue in the VSD group was lower than that in the control group at the seventh day(P<0.05).6.MASSON trichrome staining showed that:The speed of re-epithelialization in the VSD group was higher than that in the control group at the seventh day(P<0.05).Conclusion:1.Vacuum sealing drainage can accelerate wound healing in rats.2.Vacuum sealing drainage can promote the protein expression of type I collagen and type Ⅲ collagen and improve the ratio of collagen types I/III.
