Objective To observe the curative effect,safety,quality of life,and cell immune function of patients with colorectal cancer treated with Brucea javanica oil emulsion injection combined with chemotherapy,and to explore the mechanism of Brucea javanica intestinal cancer based on network pharmacology.Methods 1.Divided 70 patients with advanced colorectal cancer into experimental group according to random number table method.And 33 cases in the control group(2 cases of shedding).The control group was given standard chemotherapy,and the experimental group was treated with brucea oil emulsion injection on the basis of chemotherapy.The efficacy,immune function,adverse reactions and quality of life of the two groups were compared before and after treatment.2.Network pharmacology research method screens out the active ingredients and targets of Brucea javanica Screen the key targets of colorectal cancer and immune function in the Genecards database,construct compound-target,disease-target,compound-target-disease,and enrich the biological function and pathway network of key targets to explore The antitumor mechanism of Brucea javanica on colorectal cancer and its possible effect on immune function.Results 1.There was no significant difference in the short-term efficacy between the two groups of patients(P > 0.05);the incidence of adverse reactions(leukopenia,neutropenia,thrombocytopenia)in the experimental group was lower than that in the control group(P<0.05);after treatment,The levels of CD3+,CD4+,CD4+/CD8+ in the two groups were lower than before treatment,and the above indicators in the experimental group were significantly higher than those in the control group(P <0.05).The levels of CD8+ and NK cells in the experimental group were significantly higher than those before treatment and the control group.There was statistical significance(P <0.05);after treatment,the overall health status of both groups decreased,but the overall health status of the experimental group was significantly higher than that of the control group,and the difference was statistically significant(P <0.05).2.Network pharmacology results:(1)Brucea contains 291 targets,colorectal cancer contains 410 targets,and 257 targets are immune-related.The results of the enrichment analysis of key targets in Brucea-colorectal cancer show that in addition to colorectal cancer-related pathways,they are also enriched in prostate cancer pathway,glioma pathway,malignant melanoma pathway,bladder cancer pathway,pancreatic cancer pathway,Chronic myeloid leukemia pathway,non-small cell lung cancer pathway,colorectal cancer pathway,endometrial cancer pathway,renal cell carcinoma pathway,small cell lung cancer pathway,etc.The antitumor effect of Brucea javanica on colorectal cancer may be related to tumor signaling pathway,p53 signaling pathway,m TOR signaling pathway,apoptosis,Erb B signaling pathway and target proteins AKT1,MAPK1,CCND1,JUN,TP53.(2)The enrichment analysis of Brucea gallbladder-key immune target showed that the target involved the immune system signaling pathway:NOD-like receptor signaling pathway,Toll-like receptor signaling pathway,cytokine-cytokine receptor interaction,hematopoietic Cell line,cytoplasmic DNA sensing pathway,RIG-I-like receptor signaling pathway,FC & RI complex signaling pathway,T-cell receptor signaling pathway;involved in immune diseases: asthma,graft versus host disease,transplant rejection;neurodegenerative Disease: Alzheimer’s disease,prion disease.Conclusions 1.Brucea javanica oil emulsion injection combined with chemotherapy can improve the immune function of patients with advanced colorectal cancer,reduce the incidence of adverse reactions after chemotherapy,and improve the quality of life of patients.2.Conclusions on the antitumor effect and immune-related effects of Brucea javanica on colorectal cancer based on internet pharmacology research:(1)Brucea javanica contains 291 targets.Brucea javanica may play a role with brucetin M,luteolin,brucetin L,brucetin P,brucetin J,brucetin B,and brucetin H.,Brucein I,Brucetin D,Brucetin B,Brucetin C,Bruceol,β-Sitosterol are closely related.(2)The antitumor mechanism of Brucea javanica on colorectal cancer may be:(1)Inhibition of tumor cell proliferation and pro-apoptotic processes: The signal pathways that affect tumor cell proliferation and apoptosis in the results of this study are mainly concentrated in the p53 signal pathway and PI3 K / AKT signaling pathway,MAPK signaling pathway,m TOR signaling pathway,Erb B signaling pathway,and apoptosis.(2)Others:The results of enrichment analysis suggest that in addition to tumor pathways,they also include regulation of metabolism,cell growth,and apoptotic processes,reflecting the anti-tumor effect of Brucea javanica through multiple pathways.(3)The enrichment analysis of Brucea gallbladder pathway shows that in addition to its antitumor effect on colorectal cancer,it can also pass through the prostate cancer pathway,glioma pathway,malignant melanoma pathway,bladder cancer pathway,pancreatic cancer pathway,and chronic myeloid.The leukemia pathway,the non-small cell lung cancer pathway,the colorectal cancer pathway,the endometrial cancer pathway,the renal cell carcinoma pathway,and the small cell lung cancer pathway play a corresponding antitumor role.(4)The mechanism of the effect of Brucea javanica on immune function may include the following aspects:(1)through NOD-like receptor signaling pathway,Toll-like receptor signaling pathway,cytokine-cytokine receptor interaction,T cell receptor signaling pathway,etc.Enhance immune function or induce anti-tumor immune response through cytokines.(2)The activated immune cells secrete cytokines to further enhance the anti-tumor immune response.
