Inonotus obliquus is an edible and medicinal fungus that grows on birch trees in colder northern climates,at latitudes of 45°N–50°N.It has been widely used as a folk medicine for the treatment of cancer,hypertension and diabetes.Recently,many pharmacological effects of Inonotus obliquus have been reported.Obesity and diabetes are spreading rapidly within China and around the world,and they have become a worldwide public health problem and threaten people’s health.In this research,the toxicity of Inonotus obliquus water extracts was firstly investigated.Then,the obese and type 2 diabetes rats model were induced to evaluate the anti-obesity and anti-diabetes effects of Inonotus obliquus.And finally,the possible mechanism underlying these effects was also studied.Safety evaluation experiment included acute toxicity test,thirty-day feeding test and the mouse sperm abnormality test.In these experiments,animals were given within the max dosage(5g/kg bodyweight).During the experiments,no mice died,and the organ index and blood biochemistry analysis were also within the normal range.These results indicated that the LD50 of Inonotus obliquus is over 5g/kg and could be safety used within 5g/kg.Secondly,obese rat model was induced by feeding with high-fat diet and Inonotus obliquus extracts(IOE)were orally administered once daily for 12 weeks.After administration of IOE for 12 weeks the body weights and epididymal fat weights were significantly decreased.Moreover,IOE treatment significantly reduced serum levels of FFA,TG and TC.Real-time polymerase chain reaction(PCR)analysis further indicated that IOE treatment down-regulated the expression of genes related to lipid synthesis in the liver and adipose tissue,such as peroxisome proliferator-activated receptor γ(PPAR-γ),sterol regulatory element binding protein 1c(Srebp1c),fatty acid synthase(Fas),acetyl-Co A carboxylase(Acc),glyceraldehydes 3-phosphate acyltrans-ferase(Gpat)and diacylglycerol acyltransferase 1,2(Dgat1,2),which may be directly responsible for the reduced hepatic TG and TC levels after the IOE treatment.These observations suggested that IOE treatment can effectively ameliorate lipid accumulation,which may be achieved through inhibiting the expression of several regulators and genes involved in lipid synthesis.Finally,type 2 diabetic rat modle was induced by STZ injection and the diabetic rats were given IOE for 15 weeks.The results showed that IOE treatment could effectively control blood glucose levels,and stimulate insulin secretion of diabetic rats.Meanwhile,IOE treatment increased insulin secreation and insulin sensitivity in peripheral organs.On the other hand,IOE treatment improved glucose and lipid metabolism of diabetic rats,such as lowering blood lipid levels,increasing glucose uptake and metabolism,inhibitting gluconeogenesis.In conclution,Inonotus obliquus have good therapeutic effects on rat’s obesity and type 2 diabete.These results may lay the foundation for the research and development of relative medicine.
