Expression Of CBX7 In Hepatocellular Carcinoma And Its Effect On Prognosis

Objective: To study the expression level of pigment frame homologous protein7(CBX7)in primary hepatocellular carcinoma,and to explore the effect of CBX7 expression on the clinical prognosis of HCC.Methods: 1)Western blotting(Wes tern-Blot)was used to detect the expression of CBX7 in 1 normal liver cell line(LO2)and 7 hepatocellular carcinoma cell lines(PLC-5,QGY-7703,SMCC-7721,HepG2,Hep-3B,Huh-7,SK-1).2)the pathological specimens and clinical data of 135 pat ients with HCC treated by operation in the Department of Hepatobiliary and Pancr eatic surgery of our hospital from 2014 to 2016 were collected.The expression of CBX7 in cancer tissues and adjacent tissues was detected by immunohistochemistry.To analyze the relationship between the clinicopathological factors related to the e xpression of CBX7 and the prognostic effect of the level of CBX7 expression.Res ults: 1)the expression of CBX7 in normal hepatocyte cell line was significantly lo wer than that in hepatocellular carcinoma cell line in cellular level of WB detectio n.2)there is an imbalance in the expression of CBX7 in patients with hepatocellu lar carcinoma.Compared with the adjacent tissues,the expression of CBX7 in HC C was significantly up-regulated,mainly located in the nucleus of HCC.3)the ex pression of CBX7 was significantly correlated with the clinicopathological features of vascular tumor thrombus(rs=-0.28),degree of differentiation(rs=-0.46),MVI(rs=-0.17)and degree of liver cirrhosis(rs=0.36)in patients with HCC.4)the disease-free survival time and overall survival time of patients with high expression of C BX7 were shorter than those of patients with low expression of CBX7(P < 0.05),and CBX7 was an independent factor affecting the total survival time of patients with hepatocellular carcinoma(P < 0.05).Conclusion: CBX7 is associated with ma ny clinicopathological features of patients with hepatocellular carcinoma,and the ex pression level of CBX7 is negatively correlated with the prognosis of patients with HCC,which is an independent factor affecting the total survival time of hepatocellular carcinoma.Clinical detection of CBX7 expression may play a predictive role in predicting the overall survival time of hepatocellular carcinoma.
