ObjectiveTo explore the expression,clinical significance and functions of microRNA-142 in renal cell carcinoma.Methods1)The microRNAs expression in renal cell carcinoma were screened out by TCGA datebase.2)The expression of miR-142 in renal cell carcinoma specimens,normal tissues and renal cell carcinoma cell lines was assessed by qRT-PCR.3)miR-142 inhibitator was transfected into 786-O and Caki-1 cells.4)CCK-8 and colony formation assay were used to investigate the proliferation of renal cell carcinoma cell lines.4)The expression of APC protein levels were assessd by IHC and Western blot assay.5)Lucoferase assay was used to determin the relationship between miR-142 and 3’-UTRof APC.Results1)31cases of renal cell carcinoma and adjacent tissues were analysed in TCGA databa-se implicited that the expression of miR-142 was upregulation.2)62 cases of renal cell ca-rcinoma tissues and 30 cases of normal tissue showed significantly higher expressio-n of miR-142 in tumor tissue,and there was a correlationship between expression of miR-142 with tumor stage and tumor grade(P<0.05).3)The expression of miR-142 in 786-O and C-aki-1 cells was significant suppressed after transfection with miR-142 inhibitator(P<0.05).4)The proliferation and colony formation of 786-O and Caki-1 cells were obviously redu-ced after transfection with miR-142 inhibitator(P<0.05).5)miR-142 can regulate the ex-pression of APC protein.ConclusionUpregulation of miR-142 can promote proliferation and colony formation in renal cell carcinoma by target APC,and the expression of miR-142 was related with the grade and st-age in renal cell carcinoma.
