Objective:1.To investigate the expression of glutathione peroxidase 7(GPX7)in Papillary thyroid carcinoma(PTC),and to explore its role in the formation and progression of PTC.2.Compare the expression of GPX7 and Galectin-3 in PTC,exploring the application value of GPX7 in differentiating PTC,and the potential relationship between GPX7/Galectin-3 and tumor clinicopathologic factors.Methods:Immunohistochemistry(IHC)was used to detect the expression of GPX7 and Galectin-3 in 40 PTC and 40 normal tissues adjacent to cancer,and to explore the relationship between the expression of GPX7/Galectin-3 and clinicopathological factors.1.In PTC’s differential diagnosis,HE staining is used as the gold standard for pathological tissue judgment,and GPX7 and Galectin-3 were evaluated using diagnostic tests including accuracy/effectivity and reliability.Results:1.GPX7 is mainly expressed in the cytoplasm of thyroid cells.In PTC,GPX7 had a positive rate of 87.5%;the positive rate of GPX7 in normal tissues next to thyroid cancer was 22.5%.In PTC,GPX7 Positive rate was Obviously higher than that in normal tissues adjacent to thyroid cancer(P<0.05).In PTC,the positive rate of GPX7 was significantly different between groups with or without lymph node metastasis(P<0.05).GPX7 Positive rate in PTC was not related to other clinicopathological factors including age,gender and tumor diameter,etc(P>0.05).2.Galectin-3 is expressed in the cytoplasm and nucleus of thyroid cells.In PTC,Galectin-3 positive rate is 95%;In normal tissues close to thyroid cancer,Gal-3 is 15%positive.Galectin-3 positive rate in normal tissues adjacent to thyroid cancer was lower than that in PTC,and the difference was significant(P<0.05).And there was no significant difference between clinical pathological factors(including age,gender and tumor diameter,etc.)(P>0.05).3.In PTC,the expression relationship between GPX7 and Galectin-3 was positively correlated and statistically significant(r=0.359,P<0.05).4.For the identification of PTC,Galectin-3 has higher sensitivity,specificity,negative predictive value,positive predictive,diagnostic accuracy and Kappa value than GPX7.However,the combination of the two antibodies has higher specificity/diagnostic accuracy/and positive predictive value in the tandem test,with the highest sensitivity in parallel testing.Conclusions:1.GPX7 is mainly expressed in the cytoplasm of thyroid cells.The expression of GPX7 in PTC tissues was higher than that in normal tissues adjacent to thyroid cancer,and the positive expression rate was obviously correlated with lymph node metastasis.2.Galectin-3 is expressed in both the cytoplasm and nucleus of thyroid cells.Galectin-3 positive rate in normal tissues adjacent to thyroid cancer was significantly lower than that of PTC.3.The expression of GPX7 and Galectin-3 was positively correlated in PTC.4.In the differential diagnosis between PTC and normal tissues adjacent to cancer,simultaneous detection of GPX7 and Galectin-3 can improve the sensitivity and specificity of pathological diagnosis.
