[Objective] To detect the Expression of Galectin-3 and MMP-2 in papillary thyroid carcinoma and clinical significance[Methods] Collected thyroid resection of the tissue samples from 80 cases during April to December in 2010. According to the histopathologic diagnosis is divided into normal control group of 12 cases(distance thyroid adenoma organization 0.5 cm outside the normal tissue), benign lesions group of 40 cases(tuberous strumous 18 cases, thyroid adenoma in 12 cases, hashimoto's thyroiditis 10 cases) and papillary thyroid carcinoma group of 40 cases. Using immunohistochemistry half quantitative method respectively to detect the expression of Galectin-3 and MMP-2, statistical analysis of the expression of Galectin-3 and MMP-2 in each level difference,to identify the relationship between Galectin-3 and MMP-2 and papillary thyroid carcinoma.[Result]1.Galectin-3 and MMP-2 in normal control group do not express; the expression level of Galectin-3 and MMP-2 in benign lesions is low, the expression of Galectin-3 and MMP-2 no statistical difference in tuberous strumous, thyroid adenoma, Hashimoto's thyroiditis (P>0.05);the expression level of Galectin-3 and MMP-2 in papillary thyroid carcinoma group is significantly higher than normal controls group and benign lesions group (P<0.05); the expression level of Galectin-3 and MMP-2 between benign lesions and normal controls no statistically significant difference (P>0.05). Normal controls group, benign lesions group, thyroid papillary carcinoma group Galectin-3 positive is 0%,27.5%,85% respectively; MMP-2 positive is 0%,20%,82.5% respectively.2.The expression level of Galectin-3 and MMP-2 in papillary thyroid carcinoma, the tumor outside capsule is higher than in capsule,and with lymph node metastasis is higher than without lymph node metastasis (P<0.05);the expression level of Galectin-3 and MMP-2 in papillary thyroid carcinoma do not have relationship with age(<45 or≥45),tumor diameter(≤1cm or>1cm) and tumor distribution (single focal distribution or multifocal distribution)3.The correlation analysis between Galectin-3 and MMP-2 in papillary thyroid carcinoma was positively correlated, correlation coefficient r=0.729, P<0.05.4.With Galectin-3 as means to detect papillary thyroid carcinoma, the sensitivity, specificity, diagnostic accuracy, positive predictive value, negative predictive value and Kappa index were 85%,72.5%,78.75%,75.6%,82.86%,0.575; and with MMP-2 the figures were 82.5%,80%,81.25%,80.49%,82.05%,0.625; Galectin-3 combined with MMP-2 the figures were 77.5%,92.5%,85%,91.18%,80.43%,0.7。[Conclusion]1. The expression level of Galectin-3 and MMP-2 in papillary thyroid carcinoma group is significantly higher than normal controls and benign lesions group, and papillary thyroid carcinoma group with lymph node metastasis is higher than without lymph node metastasis; the tumor outside capsule is higher than in capsule. Both of them do not have relationship with age, tumor diameter and distribution of tumor. It is means that the expression level of Galectin-3 and MMP-2 in papillary thyroid carcinoma is very higher, especially with metastasis, Galectin-3 and MMP-2 play an important role in papillary thyroid carcinoma occurrence, development and invaded and transfer process.2. The expression of Galectin-3 and MMP-2 in papillary thyroid carcinoma was positively correlated. The high expression Galectin-3 make the expression of MMP-2 to increase. They promote each other and mutual assistance in the thyroid papillary carcinoma invasion and metastasis process.3. Galectin-3 combined with MMP-2 as means to identify the benign thyroid tumor and the malignant one. |
