Objective:To establish a mouse model of cough variant asthma(CVA),observe the effect of Huanglong Zhike oral liquid on airway inflammation in CVA model mice,and detect TLR4,MyD88 and NF-κB,further explore the possible mechanism of Huanglong Zhike oral liquid in reducing airway inflammation of CVA.Methods:Fifty BALB/c mice were randomly divided into five groups:control group,the model group,dexamethasone group and Huanglong Zhike Oral Liquid low and high dose groups,with 10 mice in each group.In addition to the control group,the other groups were modeled.The mice model of cough variant asthma was established by intraperitoneal injection of ovalbumin and aluminum hydroxide,sensitization and inhalation of ovalbumin.The mice in Huanglong Zhike Oral Liquid groups were given 0.75 g/kg and 1.5 g/kg by gavage respectively once a day,and were killed 14 days later.Serum was collected,right lung tissue was used for protein detection,and left lung tissue was used for histopathological staining.The levels of interleukin-4(IL-4),IL-5 and IL-13 were detected by ELISA.The lung tissues of mice were isolated from mice,and lung morphology was observed after HE staining;the expression of Toll like receptor 4(TLR4),myeloid differentiation factor 88(MyD88)and nuclear factor-κB(NF-κB)were detected by Western blot.Results:The levels of IL-4,IL-5 and IL-13 in the model group were significantly higher than those in the control group.HE staining showed that inflammatory cells infiltrated around the bronchi and blood vessels.Compared with the model group,the pathological sections of dexamethasone group,Huanglong Zhike Oral Liquid low-dose group and high-dose group were improved,and the levels of inflammatory cells and cytokines in serum were significantly reduced.The expression of TLR4,MyD88 and NF-κB in lung tissue of the model group was significantly higher than that of the normal group,and the difference was statistically significant(P<0.01).Compared with the model group,the expression of TLR4,MyD88 and NF-κB in Huanglong Zhike Oral Liquid group decreased significantly(P<0.01).Conclusions:1.Huanglong Zhike Oral Liquid can improve airway inflammation and airway hyperresponsiveness in cough variant asthma model mice.2.TLR4/MyD88/NF-κB signaling pathway is involved in airway inflammation in cough variant asthma mice.3.Huanglong Zhike Oral Liquid can regulate the release of inflammatory cytokines through the TLR4/MyD88/NF-κB signal pathway,so as to regulate airway inflammation in mice and improve cough variant asthma. |
