| ObjectiveGlioma is the most common primary intracranial tumor.At present,for the treatment of gliomas,the first choice is still to resect the tumor tissue as much as possible by microneurosurgery,followed by radiotherapy and chemotherapy.However,under the current treatment mode,glioma is "expensive,easy to relapse,poor prognosis".Previous studies have reported that neurodevelopmental down regulated 8(NEDD8)and chemokine like factor superfamily 6(chemokine like factor superfamily 6,CMTM6)is highly expressed in lung cancer,liver cancer,breast cancer,ovarian cancer,melanoma and other human tumors.NEDD8 and CMTM6 may participate in the process of tumorigenesis and development as important factors.However,there are few studies on the expression of NEDD8 and CMTM6 in glioma.In view of this,immunohistochemistry and Western blot were used in this study The expression of NEDD8 and CMTM6 in gliomas was detected by blot.The difference between the expression of NEDD8 and CMTM6 in gliomas and control brain was analyzed by statistical method.Whether there was correlation between the expression of NEDD8 and CMTM6 in gliomas,and the relationship between the expression of NEDD8 and CMTM6 and the clinicopathological parameters of gliomas,and the influence on the prognosis of gliomas,so as to further understand the gum The molecular pathogenesis of tumor and the clinical treatment of glioma provide some reference.MethodsFrom January 1,2013 to January 1,2016,tissue samples of 80 glioma patients who received surgical treatment in neurosurgery of the Fifth Affiliated Hospital of Zhengzhou University and 30 craniocerebral trauma patients who underwent"decompressive craniectomy" in neurosurgery of the Fifth Affiliated Hospital of Zhengzhou University were selected.Inclusion criteria:first onset;definite intracranial space occupying lesions in imaging(CT,MRI),and at least two experienced neurosurgeons judged as gliomas;received surgical treatment in neurosurgery of the Fifth Affiliated Hospital of Zhengzhou University;imaging(CT,MRI)showed that the tumor was located in frontal lobe,temporal lobe and parietal lobe;no preoperative radiotherapy,chemotherapy and other treatment history;postoperative Patients or their families signed informed consent to participate in the study.Among the 80 patients,53 were high pathological grade gliomas(WHOⅢ-Ⅳ)and 27 were low pathological grade gliomas(WHOⅠ-Ⅱ).The expression of NEDD8 and CMTM6 in gliomas and normal control tissues was detected by immunohistochemistry and Western blotting.The difference of expression of NEDD8 and CMTM6 was analyzed by χ2 test and two independent samples t-test.The correlation of expression of NEDD8 and CMTM6 in gliomas was analyzed by Spearman correlation analysis.The expression of NEDD8 and CMTM6 was analyzed by χ2 test and several clinicopathological features were included The relationship between the parameters(age,gender,tumor diameter,tumor location,preoperative epilepsy,preoperative KPS score,preoperative dizziness,tumor cell type,pathological grade of gliomas),the relationship between the expression of NEDD8 and CMTM6 in gliomas and the prognosis of patients was detected by Kaplan Meier method,and Cox regression multivariate model was established to analyze the independent risk factors affecting the prognosis of patients.Results1.The positive expression rate of NEDD8 and CMTM6 in gliomas was significantly higher than that in control brain(P<0.05).The positive expression rate of NEDD8 and CMTM6 in high pathological grade gliomas was significantly higher than that in low pathological grade gliomas(P<0.05);2.There was a positive correlation between the expression of NEDD8 and CMTM6 in gliomas(r=0.458,P<0.001);3.The expression of NEDD8 in gliomas was related to tumor diameter,preoperative KPS score and pathological grade of gliomas(P<0.05);the expression of CMTM6 in gliomas was related to patients’ age,preoperative epilepsy,preoperative dizziness history and pathological grade of gliomas(P<0.05);4.The higher the expression level of NEDD8 and CMTM6 in gliomas,the shorter the survival time and the worse the prognosis.The prognosis of gliomas with high expression of NEDD8 and CMTM6 was worse(P<0.05);5.The expression level of NEDD8 in glioma,CMTM6 in glioma and pathological grade of glioma were independent risk factors(P<0.05).Conclusion1.The high expression of NEDD8 and CMTM6 in gliomas increased with the increase of pathological grade of gliomas,suggesting that NEDD8 and CMTM6 may be involved in the occurrence and development of gliomas as important factors,and NEDD8 and CMTM6 may be used to judge the biological characteristics of gliomas.2.There is a positive correlation between the expression of NEDD8 and CMTM6 in gliomas,suggesting that there may be a common pathway between them,which can be further studied,which is very helpful to explore the pathogenesis of gliomas related molecules;3.The expression level of NEDD8 and CMTM6 in gliomas is negatively correlated with the prognosis of patients.The expression level of NEDD8 in gliomas,the expression level of CMTM6 in gliomas and the pathological grade of gliomas are independent risk factors for the prognosis of gliomas.NEDD8 and CMTM6 are expected to be used to evaluate the prognosis of gliomas. |
