ObjectiveThe object of this study is obese rats,EA at points of different body parts:Zusanli(ST36)?Tianshu(ST25)?Quchi(LI11),observe the differences in body weight,epididymal white adipose tissue(eWAT),and liver of rats before and after intervention,and using neuroelectrophysiological methods to record the state of sympathetic activity on different rats,in order to clarify the difference in weight loss of EA at different parts and its possible neurobiological mechanism,and provide scientific basis for clinical application.MethodsThe diet-induced obesity(DIO)rat model was made through high-fat diet,after successful modeling,the rats were randomly divided into DIO model group,and EA(2/15Hz?2mA?2min)group(divided into 3 subgroups:ST36?ST25?and LI11 Group),treatment was performed once daily for 6 days/week,and for 4 weeks.After the treatment,observe the differences in obesity-related indicators(body weight of rats,eWAT wet weight,and liver wet weight),measure and record the inguinal white adipose tissue(iWAT)on the sympathetic nerve of the rat through neuroelectrophysiological techniques.After it was stabilized,observe the effect of EA(2/15 Hz,2 mA,2 min)on the frequency of sympathetic nerve discharge in ST36/ST25/LI11(acupoints are on the same side of the operation).Results1.The body mass of the DIO model group was significantly higher than that of the normal group,After 4 weeks of EA,the weight of rats in EA groups(including ST36,ST25,and LI11 group)was significantly lower than that in the DIO model group(P<0.05),and the ST25 group inhibited weight gain most significantly(weight:X normal:549.13g;XDIO model:643.28g;X ST36:592.26g;X ST25:569.26g;X LI11:592.83g);The percentage of weight gain in EA group(including ST36,ST25,and LI11 group)were significantly lower than that in the normal and the DIO model group(P<0.05),and the changes of weight gain of ST25 group were significantly lower than those of LI11 group and ST36 group;2.After 4 weeks of EA,the wet weight of eWAT and liver in the EA groups(including ST36,ST25,and LI11 group)were significantly lower than those in the DIO model group,and there was a statistically significant difference of eWAT weight and liver weight between the ST25 group?ST36 group and the DIO model group(P<0.05)(weight of eWAT:X DIO model:13.65g;XST36:8.20g;X ST25:7.16g;XLI11:10.72g;weight of liver:X DIO model:16.03g;X ST36:13.73g;X ST25:13.66g;XLI11:14.62g);3.The spontaneous discharge frequency of iWAT sympathetic nerves in DIO model group and EA group are significantly lower than that in normal group(discharge frequency:X normal:5.13Hz;XDIO model:0.31Hz;X ST36:0.34Hz;X ST25:0.77Hz;XLI11:0.63Hz);Effective rate of EA at different acupoints on iWAT sympathetic nerve discharge(including excitation and suppression)is ST36>ST25>LI11 in order from high to low;And the effect of electricity on iMAT sympathetic nerve discharge in each group is mainly excited(hereinafter referred to as acupuncture excitability rate),the excitability rate was highest when EA at ST36 in the normal group,and ST25 in the DIO model group,fter electroacupuncture intervention,ST36 group had the highest acupuncture excitement rate.Conclusion1.Acupoints in different parts of the EA have good weight loss effects,but there are differences(The effect of the ST25 in abdomen is the best,followed by ST36,the effect of LI11is relatively weak);2.Both EA at the ST25 in abdomen and ST36 at the lower extremity have a good effect of reducing fat,and the effect of LI11 in upper limb is weak;3..The iWAT sympathetic nerve sensitivity of rats decreased after obesity(including the DIO model group and the EA groups),and the frequency of spontaneous discharge was significantly reduced.The spontaneous firing of iWAT sympathetic nerves of rats after EA treatment increased compared with the obese state;4..EA at different acupoints can activate the iWAT sympathetic nerve,and there is a significant acupoint specificity(the state of the body affects the amount of acupoint effects,ST36 is the highest in the normal,while ST25 is the highest in the obese,the effect of ST36 is largest when the body returns to normal after losing weight).In summary,this study shows that:clinically effective and representative acupoints for weight loss,the effect is determined by the difference in innervation and the difference in body state,which provides part of the biological basis for the clinical science to choose weight loss programs rationally.
