Objective:To investigate the correlation between alcohol dependent patients and gene polymorphisms of hla-dra,HTR7,VMAT2 and ADH1C in Chinese han male populationObjects and methods:Select 230 cases in line with the diagnostic criteria of ICD-10 han patients with alcohol dependence as experimental group,collect healthy controls of 230 cases by using polymerase chain reaction(PCR)and sequences Sanger sequencing methods of 230 patients with alcohol dependence and 230 cases of healthy controls HLA-on DRA gene loci rs4935356,HTR7 gene loci rs 7904560,VMAT2 gene loci rs 363387,ADH1C gene loci rs1614972 genotype and allele frequency were determined.SPSS17.0 Chinese version software was used for statistical processing of experimental data,independent sample t test was used for data processing between groups,Hardy-Weinberg genetic balance test was used for sample genotypes,chi-square test or Fisher exact probability method was used for genotype composition ratio and allele frequency distribution difference comparison.The significant level was=0.05.Results:1.Clinical study(1)the average age of the patients with alcohol dependence was(46.465±9.618)years old,and the average age of the healthy control group was(45.061±12.659)years old,and there was no significant difference between the two groups in age(t=1.340,P=0.181).(2)on MAST score,there were significant differences(t=54.614,P=0.000)between 230 cases of alcohol-dependent patients(29.948±7.989)and 230 healthy controls(1.057±0.731).2.Analysis of gene sequencing results of alcohol-dependent patients and healthy control group(1)there was no statistical difference in the genotype composition of HLA-DRA SNP site rs4935356 between the alcohol-dependent patients and the healthy control group(χ~2=5.856,P=0.210),nor in the frequency distribution of allele between the alcohol-dependent patients and the healthy control group(χ~2=5.74,P=0.057).(2)the difference in allele frequency distribution of the SNP site rs 7904560 of HTR7 in alcohol-dependent patients was statistically different from that in healthy controls(χ~2=14.625,P=0.010),and the difference in allele frequency distribution was also statistically different from that in healthy controls(X~2=33.510,P=0.000),and the A allele might be the susceptible gene.(3)the genotype composition ratio of VMAT2 SNP site rs 363387 in alcohol-dependent patients was statistically different from that in healthy controls(-,*P=0.041),but the allele frequency distribution difference was not statistically different from that in healthy controls(χ~2=0.829,P=0.363).(4)of the patients with alcohol dependence of ADH1C rs1614972 SNP locus of genotype and allele frequency distribution difference compared with healthy control group statistically difference(χ~2=19.271,P=0.000)and allele frequency distribution difference compared with healthy controls also statistically difference(χ~2=28.195,P=0.000),prompted G allele may be the susceptibility genes.Conclusion:in Chinese han male population(1)the polymorphism of ADH1C rs1614972 may be related to alcohol dependence,and the G allele may be its susceptibility gene;(2)the polymorphism of HTR7 rs7904560 May be related to alcohol dependence,and the A allele may be its susceptibility gene;(3)the polymorphism of VMAT2 rs 363387 May be related to alcohol dependence.(4)polymorphism of HLA-DRA rs4935356 may be independent of alcohol dependence.
