| Objective : Through research Yiqi Tongluo anti-cancer recipe coalition for tegafur,gimeracil and oteracil potassium(TGO)medicated serum on gastric cancer SGC-7901 cell proliferation and cell cycle,reveal their combined chemotherapy in the treatment of gastric cancer may mechanism of action,for clinical treatment malignant tumor of alimentary canal such as gastric cancer to provide new method and powerful experimental basis.Methods: 1 According to the serum pharmacology method,it is divided into the TGO group,the Yiqi Tongluo anti-cancer recipe combined with TGO group,and 0.9% Na Cl solution group used to prepare the medicated serum;2 The proliferation of SGC-7901 cells was measured by CCK-8 method;3 The cell circle was measured by FCM analysis;4 Western Blot was used to measure the expression levels of cyclin A、cyclin D1、cyclin E and CDK2.Results: 1 Except the normal saline group,the drug-containing serum of each group inhibited the proliferation of SGC-7901 cells,with the highest inhibition rate of 17.74% in the 72 h TCM combined with TGO group(P<0.05).2 Compared with the normal saline group,the proportion of G2/M phase cells in the drug-containing serum SGC-7901 in each group decreased to different degrees(P<0.05).3 The combination of TCM and sergio can reduce the expression of cyclin A in SGC-7901 cells(P<0.05)and increase the expression of CDK2(P<0.05),while the expression of cyclin D1 and cyclin E has no statistical significance(P > 0.05).Conclusions: 1 Yiqi Tongluo anti-cancer prescription and TGO can effectively inhibit the proliferation of SGC-7901,and the inhitory of two drugs is better than the single drug.2 Yiqi Tongluo anti-cancer prescription and TGO can effectively inhibit the S/G2 phase transformation,and the inhitory of two drugs is better than the single drug.3 Down-regulated expression of the protein cyclin A may be one of the mechanisms by which them exert S/G2 phase transformation inhibition on sgc-7901 cells. |
