| Objective: Routine tumor treatment methods include surgical excision,chemotherapy and radiation,which have the characteristics of great minus effects and high-cost.Arsenical also has considerable side effects in the treatment of tumors through intravenous or oral administration.Iontophoresis of Chinese medicine is a unique method of combining Chinese medicine and current physics,with a transfer process of ionized drug molecules through the skin biofilm under the influence of electric current.It is a method of using direct current to transfer ionic drugs into the skin and into tissue or fluid circulation by electrodes.Realgar,one of the most common mineral drugs,has anti-tumor effect.The combination of iontophoresis and realgar anti-tumor effect can greatly reduce the side effects on the body.In order to investigate the effect of electric iontophoresis on the anti-tumor effect of Chinese medicine realgar,the experiment was carried out in this paper.Methods: Acidithiobacillus ferrooxidans ATCC23270 was used for the biochemical oxidation of realgar insoluble in water.The active component such as arsenic was effectively released into an aqueous solution and,obtained realgar microbiological leaching solution.Then proliferation inhibition experiments of human breast cancer cell line T47 D in vitro was performed.A mouse tumor model was carried out through iontophoresis experiments.Results: It has a significant inhibitory effect on the 0.5-2.0μg/mL concentration in the experimental results of in vitro cell proliferation inhibition test,induced apoptosis,and has a strong time dependence.The treatment of iontophoresis of realgar bioleaching solution has a strong tumor growth inhibitory effect on T47 D breast cancer cells in mice model,and with dose and time dependent.The high dose group(2mg/mL)was statistically significant compared with the control group in tumor size(P < 0.05).Conclusion: The realgar bioleaching solution could induce apoptosis in human breast cancer cell line T47 D in vitro.Iontophoresis of realgar leaching solution had tumor inhibitory effect on T47 D mice model. |
