C. elegans is the simplest differentiated organism with a short life cycle, small enough to be handled in large numbers, easy to be cultivated. Since then, C. elegans has been a powerful tool to provide insights into the molecular mechanisms underlying numerous biological processes, such as programed cell death, cell cycle, sex determination etc. The adult C. elegans hermaphrodite is about 1 mm long and 80um in diameter. Animals can be grown on a diet of Escherichia coli on agar plates or in liquid medium. Its life cycle is short (3.5 days at 20°C) and each hermaphrodite produces -300 larvae by self-fertilization. Crossfertilization with occasional male individual facilitates genetic analysis. C. elegans is becoming a popular platform that can be used to gain a deeper understanding of the mechanism of action of various human drugs, as well as for the discovery of new compounds. In our experiment, we used C. elegans as a tool for the identification of antibacterial activity of reagar bioleaching solution. Also we used it as a tool for the measurement of anti-cancer activity of reagar bioleaching solution. Data from mice in our lab showed that reagar bioleaching solution could inhibit some pathogens, also it could inhibit soild tumor. Similar results can be obtained from our experiment.Data from this experiment show, the concentration of 0.12μg/ml of realgar bioleaching solution can effectively inhibit the infection of Enterococcus faecalis,6.40μg/ml of Realgar bioleaching solution can suppress formation of the Multivulv. The toxicity results indicate that realgar reagar bioleaching solutions in similar dose lead to different half lethal death time for different strains of C.elegans. half lethal death times are from 7 hours to 12 hours,Our results suggest that it is practical and efficient to use C. elegans as a model organism to discover both antibacterial and anticancer drug candidates. |
