Objective To investigate the protective effect of Buqing Granule on diabetic cataract lens in db/db mice,and to elucidate the molecular mechanism of Buqing Granule in delaying the development of diabetic cataract lens in db/db mice.Methods db/db mice were selected to simulate type II diabetic cataract.The mice were given intragastrically administrated with Buqing granules to observe the degree of lens opacity in mice.HE staining was used to detect the pathological changes of db/db mouse lens.Immunohistochemistry The expression of Beclin1,Bcl-2 and mTOR protein in the lens of mice was detected.The model of oxidative damage of human lens epithelial cells(HLE)was induced by H2O2,and the serum containing Buqing granules was used to intervene.The drug-containing serum of Buqing granules was detected by CCK8 method.Wound healing experiments were performed to examine the effects of migration abilities,and flow cytometry was used to examine the effects of cell cycle.The expression of mTOR,AMPK,PI3K,Akt protein and gene in HLE cells was detected by Western blot and RT-PCR.Results In vitro observation of db/db mouse lens showed that the lens opacity of mice in the Buqing granule group was lighter than that of the model group.HE staining showed that the lens lesions of the mice in the Buqing granule group were lighter than those in the model group,and the CCK8 results showed a patch The cell viability of the granule group was higher than that of the model group;the scratch test results showed that the cell migration ability of the granule group was stronger than that of the model group;the flow cytometry showed that the proportion of cells in the G2 and S phase of the granule was higher than that of the model group;WB and RT-The PCR results showed that compared with the model group,the expression of AMPK and Beclin1 was increased,and the expression of PI3K,Akt,mTOR and Bcl-2 protein was decreased.Conclusion:Bu Qing Granules can up-regulate the expression of AMPK protein,down-regulate the expression of PI3K and Akt in mouse lens,activate AMPK-mTOR signaling pathway,inhibit PI3K-Akt-mTOR signaling pathway,promote autophagy of db/db mouse lens,and delay lens turbid.
