ObjectiveTo investigate the effect of Yanghe Decoction on inflammatory factors of synovial tissue induced by papain in rabbit knee osteoarthritis model,in order to further clarify the mechanism of Yanghe Decoction(YHD)in treating osteoarthritis.MethodsForty-four healthy New Zealand rabbits aged 4-6 months were randomly divided into a blank control group(n=9)and a model group(n=33)according to the random number table method.The blank control group was not modeled,and the other animals were modeled.The knee osteoarthritis model was prepared by intra-articular injection of 4% papain solution.Two weeks later,three randomly selected models from the blank control group and the model group were used for model identification.Newly-developed New Zealand white rabbits were randomly divided into model group,YHD low dose group,YHD medium dose group,YHD high dose group and positive control group,with 6 rats in each group.After the successful establishment of the model,intervention therapy was started.Each rabbit in the low,medium and high doses of YHD was given 3 g/kg·d,6 g/kg·d,12 g/kg·d YHD,each rabbit in positive control group was given 24 mg/kg·d Celebrex.The blank control group and the model group were given the same amount of normal saline.Once a day,continuous gavage treatment for 2 weeks.After 2 weeks of treatment,the knee joint swelling degree was observed,the knee joint circumference and local skin temperature were measured,the anesthesia was taken,the knee joint was observed,the rabbit knee joint synovium was taken,the conventional fixed embedding and sectioning,and the section was subjected to hematoxylin and eosin staining.Lequesne score was used to evaluate animal behavior,enzyme-linked immunosorbent assay for detection of IL-1β,TNF-α,COX-2,PGE2,NO content in serum and joint fluid,microscopic observation of synovial tissue pathology;immunohistochemistry and quantitative analysis of images IL-1β,TNF-α,COX-2,iNOS expression.Results1.Knee joint skin temperature and joint circumference results: Compared with the blank group,the skin temperature of the knee joint in the model group was increased(P<0.05),and the circumference of the joint was significantly increased(P<0.05).Compared with the model group,each treatment the skin temperature of the knee joint decreased(P<0.05),and the circumference of the joint decreased(P<0.05).Compared with the positive control group,the local skin temperature of the Yanghetang group was lower(P<0.05),and the joint circumference was smaller(P<0.05).2.Results of Lequesne score: After the model was established,the score of the model group was higher than that of the blank control group(P<0.05).Compared with the model group,the scores of the positive and negative control groups of the Yanghe Decoction group and the positive control group were decreased(P< 0.05);Compared with the positive control group,the Yanghetang score was lower,but there was no significant difference between the two(P>0.05).3.ELISA test results: Compared with the blank control group,the levels of IL-1β,TNF-α,COX-2,PGE2 and NO in the serum and joint fluid of the model group increased significantly(P<0.05);compared with the model group,Yanghe the contents of IL-1β,TNF-α,COX-2,PGE2 and NO in serum and joint fluid of the middle dose group and the positive control group were decreased(P<0.05).4.HE staining results: Compared with the blank group,the model group showed significant synovial cell proliferation,cell arrangement disorder,more lymphocytic infiltration and a small amount of neutrophils,vascular proliferation,hyperemia,fibrous tissue hyperplasia;After treatment,compared with the model group,the synovial proliferative layer,lymphocyte neutrophil infiltration and vascular proliferation were improved in each treatment group.Among them,the Yang and Tangzhong dose group and the positive control group were significantly reduced in proliferation,inflammatory cells.cut back.Immunohistochemical results: Compared with the blank control group,the expression of IL-1β,TNF-α,COX-2 and iNOS in the model group was increased,and the IOD value was increased(P<0.05).Compared with the model group.In the Yang and Tangzhong dose group and the positive control group,the expression of IL-1β,TNF-αCOX-2 and iNOS in the synovial membrane was decreased,and the IOD value was decreased(P<0.05).ConclusionThis experimental study found that Yanghe Decoction can alleviate joint swelling and skin temperature in rabbit knee osteoarthritis model,improve knee joint function,by inhibiting COX-2 inflammatory pathway and iNOS inflammatory pathway,the expression of IL-1β、TNF-α,COX-2,PGE2 and iNOS alleviated the symptoms of synovial inflammation in rabbit knee osteoarthritis and delayed the progression of knee osteoarthritis.
