| Objective:To observe the influence of Zhuangjinyangxue decoction on the model of rabbit osteoarthritis, and explore the mechanisl in the treatment of rabbit osteoarthritis. Then we can provid the theoretical basis for clinical treatment and further study.Methods:24 healthy New Zealand rabbit were randomly divided into blank group(n=8), model group(n=8) and Zhuangjinyangxue group(n=8). Model group and Zhuangjnyangxue group in accordance with the modified Hulth model technique. Zhuangjinyangxue group were given Zhuangjinyangxue decoction, and blank group and model group were given the same amount of physiological saline, twice a day, curing for 8 week seontinuously. The content of blood serum and synovial fluid of NO, IL-1 detenmed by ELISA. The content of arthroidal cartilage of iNOS detenmed by Western blot.Observing the pathological changes of arthroidal cartilage and being graded by Mankin’s method.Results:1. Observing the pathological changes of arthroidal cartilage in the naked eye and optical microscope:the arthroidal cartilage of blank group was smooth and intact. Compared to blank group, the arthroidal cartilage was damaged obviously,the surface of articular cartilage was defective, rough and fissured, four layer structure of cartilage is blurry, the amount of cartilage cells decreased significantly, the distribution of cartilage cells were inhomogenous, a lot of undifferentiated cartilage cells aggregate in the middle layer, the tidemark was incomplete. Zhuangjinyangxue group between blank group and model group, have improved significantly than the model group, and reduce the damage of arthroidal cartilage.The Mankin’s score showed:the Mankin’s score of model group compared with blank group increased significantly, there was significant difference(P<0.01); the Mankin’s score of Zhuangjinyangxue group compared with mode group is different(P<0.05), the Mankin’s score decreased,there was statistically difference.2. The content of NO, IL-1 in blood serum and synovial fluid showed:The content of blood serum and synovial fluid of NO, IL-1 in model group compared with blank group increased significantly(P<0.01 or P<0.05);the content of blood serum and synovial fluid of NO, IL-1 in Zhuangjinyangxue group compared with model group decreased significantly(P<0.01).3. The content of iNOS in arthroidal cartilage showed:the expression of iNOS in model group compared with blank group increased significantly(P<0.01); The expression of iNOS in Zhuangjinyangxue group compared with model group decreased significantly(P<0.01).Conclusions:1. The modified Hulth model technique can successfully establish the model of rabbit osteoarthritis;2. Zhuangjinyangxue decoction can reduce the content of NO, IL-1 in blood serum and synovial fluid effectively, and reduce the expression of iNOS in arthroidal cartilage effectively;3. Though reducing the content of NO, IL-1 and the expression of iNOS, Zhuangjinyangxue decoction can reduce the destruction of arthroidal cartilage, promote arthroidal cartilage repair, postpone the degeneration of arthroidal cartilage, relieve effectively clinical symptoms,then play a effect of prevention and treatment of OA. |
