Objective In this study,we examined the polymorphism of Klotho protein G-395 A locus and general blood biochemical indicators in Han population in Shaanxi,comparing the distribution of A G loci genotypes for gene frequencies in patients with coronary heart disease and Frank’s sign.The aim of this study was to clarify the distribution of Klotho gene polymorphism in the development of coronary heart disease and Frank’s sign.Methods From November 2017 to August 2018,209 inpatients aged 18 to 70 in the Department of Cardiology of Shaanxi People’s Hospital were selected.All of them were Han nationality and accepted coronary angiography voluntarily.According to the criteria,the patients were divided into coronary heart disease group(167 cases)and non-coronary heart disease group(42 cases),Frank’s sign positive group(70 cases)and Frank’s sign negative group(139 cases).General information,medical history and related blood biochemical examination were collected.Peripheral venous blood samples were collected by fasting centrifugation.Allele-specific primers were used to detect the genotype of Klotho gene G395-A polymorphism site by PCR,and Taq Man allele resolution method was used to genotype.SPSS software was used to analyze and compare the relationship between alleles and coronary heart disease and Frank’s sign.Result1.The subjects were divided into two groups by coronary heart disease and Frank’s sign respectively.The baseline data of the two groups were compared.Only coronary heart disease was significantly correlated with Frank’s sign(P = 0.009,P = 0.002,< 0.01).2.Among all the subjects in this study,G-395 A of Klotho gene detected GG,GA and AA genotypes.3.The genotype and allele frequencies of Klotho protein G-395 A in CHD group and non-CHD group did not conform to Hardy-Weinberg equilibrium rule(P=0.026,< 0.05),it were not representative of the population.4.The genotype and allele frequencies of Klotho protein G-395 A in Frank-positive group and Frank-negative group conform to Hardy-Weinberg equilibrium rule(P>0.05),which is representative of the population.5.The frequencies of G-395 A and alleles were 51.6,OR=6.71 and P=0.000,< 0.01 by chi-square test between Frank-positive group and Frank-negative group.The same results were obtained by using the other two models(P=0.000,<0.01),indicating that a significant relationship between G-395 A allele,and Frank’s sign positive,especially genotype A,.7.Logistic regression analysis was used to compare G-395 A genotype and clinical indicators between Frank’s sign positive group and Frank’s sign negative group.Age(P =0.02,< 0.05),allele A(P = 0.000,< 0.01)were correlated with Frank’s sign.Conclusion1.Frank’s sign positive is associated with coronary heart disease.2.Frank’s sign is related to age.3.Alleles A and G at G-395 A of Klotho gene are not correlated with coronary heart disease.4.Allele A at G-395 A of Klotho gene may be a susceptible genotype of Frank’s sign.
