Objective:To investigate the characteristics and rules on the expression of mucin genes(MUC2,MUC5AC,MUC5B,MUC6)in the sinus mucosa of patients with Chronic rhinosinusitis(CRS)and normal people,then to discuss its relationship with the severity of Chronic rhinosinusitis.Methods:34 CRS patients,who hospitalized in the department of otolaryngology,sichuan provincial people’s hospital from January 2018 to July 2018,were selected as the case groups(23 patients with nasal polyp and 11 patients without nasal polyp),and 13 patients with simple nasal septum deviation hospitalized during the same period were selected as the normal control group.The mucosa of the ostiomeatal complex was taken as the specimens during the operation.The expressions of MUC2,MUC5AC,MUC5B and MUC6 mRNA in the sinus mucosa of each group were detected by real-time fluorescence quantitative PCR(qrt-pcr).Evaluating patients’subjective illness severity by patients’Visual analogue scale(VAS)total scores of nasal obstruction,nasal discharge,head and facial pain,reduction in sense of smell.Eevaluating patients’objective illness severity by Lund-Mackay scores of the sinus CT scan during the hospitalization.SPSS23.0 software was used for statistical analysis of the experimental data.Results:1.The expression of MUC2,MUC5AC,MUC5B and MUC6 mRNA were detected in the sinus mucosa of the patients in the case group and the control group,MUC5B and MUC5AC mainly.The expression of MUC5AC and MUC5B mRNA in the case group were significantly higher than those in the normal control group,and the difference was statistically significant(P<0.05),while there was no statistically significant difference between the CRSwNP group and CRSsNP group(P>0.05).The expression of MUC2 and MUC6 mRNA between the case group and the normal control group was no statistical difference(P>0.05).2.In three groups,expression of MUC2 mRNA in sinus mucosa was positively correlated with that of MUC5AC(P<0.01).3.In the CRSwNP group,the expression of MUC5B mRNA was positively correlated with patients’preoperative VAS total scores(r_s=0.527,P<0.05).However,in the CRSsNP group,the expression of MUC5B mRNA was no significantly correlated with patients’preoperative VAS total scores(P>0.05).4.In the CRS groups,the expression of mucin genes was no significantly correlated with the pre-operative CT Lund-Mackay scores(P>0.05).Conclusion:1.There were MUC2,MUC5AC,MUC5B and MUC6 mRNA expressed in the sinus mucosa,MUC5B and MUC5AC mainly;The high expressions of MUC5AC and MUC5B mRNA may play an important role in the excessive secretion of mucus in CRS;2.There is a correlation between the expression regulation of MUC2 and MUC5AC mRNA;3.The expression of MUC5B mRNA could be used to assess the severity of the disease in patients with CRSwNP.
