ObjectiveThe therapeutic mechanism of Tai Shan Pan Shi San on abortion was studied by using Tai Shan Pan Shi San to treat abortion model rats,which provided further theoretical basis for the clinical application of Tai Shan Pan Shi San.Methods1.SPF grade SD rats(body weight 250±10 g)were used,and after 7 days of adaptive feeding,they were caged according to the random number table method.After the vaginal plug was observed,it was recorded as the first day of pregnancy,and then randomly divided into normal group,model group and treatment group.Abortion model was established by subcutaneous injection of bromocriptine solution 1mL/kg/d(concentration 0.4 mg/mL;solvent 75 % ethanol)on the 6th to 8th day of pregnancy.2.On the 1st to 11 th day of pregnancy,treatment with Taishan Lanshi Powder Decoction 10ml/kg/d.The normal group was given the same amount of normal saline and subcutaneous injection of 75 % ethanol.The model group was given the same amount of normal saline On the 12 th day of pregnancy,20 % urethane solution(5mL/kg)was intraperitoneally injected to anesthetize the pregnant rats to obtain tissues.The serum was taken out and allowed to stand for 2 h,then centrifuged and stored in a-80 ° C refrigerator.Cut the uterus tissue in pregnancy and put it in 4 % paraformaldehyde for 36 hours,then embedding.3.Determination of estradiol E2 and progesterone P in serum by ELISA.Morphological changes of decidua in uterus of maternal-fetal interface in pregnant rats by HE method.Immunohistochemical staining for expression of apoptosis-related proteins TNFR1,FADD,Caspase-8 and Caspase-3 in decidua of maternal-fetal interface in pregnant rats.The apoptosis of decidual tissue in the uterus of the maternal-fetal interface was detected by TUNEL method.Results1.The results of HE: Tai Shan Pan Shi San can significantly improve the decidual tissue structure of the uterine side in the maternal-fetal interface of pregnant rats,and the perivascular improvement is obvious.2.The results of ELISA : Tai Shan Pan Shi San can improve the content of estradiol E2 and progesterone P in the serum of pregnant rats with abortion model was increased,and E2 increased significantly.3.The results of immunohistochemistry : Tai Shan Pan Shi San can reduce the expression of TNFR1,FADD,Caspase-8 and Caspase-3 in the decidua tissue of the uterine side in the maternal-fetal interface of pregnant rats,and the most obvious decrease around the blood vessels.4.The results of TUNEL : Tai Shan Pan Shi San can reduce the apoptosis of the decidual tissue in of the maternal-fetal interface of pregnant rats,especially the apoptosis of perivascular cells.Conclusion1.Tai Shan Pan Shi San has obvious therapeutic effect on pregnant rats with abortion,which can significantly increase the content of estradiol E2 and progesterone P in the serum of pregnant rats.2.Tai Shan pan Shi San can play a role in the treatment of abortion by reducing the expression of TNFα-TNFR1 signaling pathway-related apoptotic proteins in the decidual tissue of the uterine side in the maternal-fetal interface of pregnant rats,especially perivascular expression.
