| PART ONE Study on the role of matrix metalloproteinase in missed abortion and its regulation mechanismObjective:To investigate the immune factors in missed abortion and study the role of matrix metalloproteinase in missed abortion and its regulatory mechanism.Methods:30 patients with missed abortion were selected as the research group,30 healthy pregnant women in the same period were selected as the control group.The villus of the two groups were obtained during the implementation of curettage under sterile conditions.Physiological saline treated with diethyl pyrophosphate was used to remove mucus and blood of villus.And filter paper was used to remove surface moisture.The villus obtained after removing the decidual tissue were placed in tubes that inactivated RNase and stored at-80 °C for cryopreservation.RT-PCR was used to detect the expression of NF-κB,TGF-β1,TIMP-1,MMP-9 and other related genes in villus tissues of two groups.The target genes were amplified and analyzed.Western Blot was used to detect the protein expression of NF-κB,TGF-β1,TIMP-1 and MMP-9 in the villi of patients.Results:(1)The level of TGF-β1 in plasma of the research group was lower than that in the control group(P<0.05).(2)The m RNA expression of NF-κB1,TGF-β1 and TIMP-1 in villi of the research group were lower than that in the control group,and the expression of MMP-9 m RNA was higher than that of the control group(P<0.05).(3)The level of MMP-9 protein in villi was higher than that in the control group,and the level of NF-κB p50,TGF-β1 and TIMP-1 protein in villi was lower than that in the control group(P<0.05).Conclusion:The decrease of NF-κB can affect the expression of TGF-β1,and it may influence the balance of MMP-9/TIMP-1 and lead to the excessive degradation of extracellular matrix,and result in further failure of embryo implantation and impaired placenta formation,eventually cause the termination of embryo development.PART TWO Study on the role of matrix metalloproteinase in recurrent spontaneous abortion and its regulation mechanismObjective:To investigate the expression of matrix metalloproteinase in villi of patients with recurrent spontaneous abortion,analyze the relationship and clinical significance between MMPs and RSA.Methods:24 patients with RSA were selected as the research group,26 healthy pregnant women in the same period were selected as the control group.The expression of m RNA and related proteins in the MMPs,NF-κB,IκB,and the serum TNF-α and IL-1β were detected.Results:(1)The level of TNF-α in plasma of the research group[(2.84±0.62)ug/L] was higher than that in the control group[(1.32±0.43)ug/L](P<0.05).(2)The m RNA expression of TNF-α,NF-κB and MMP-9 m RNA in villi of the research group was higher than that in the control group,and the expression of IκBβ m RNA was lower than that of the control group(P<0.05).(3)The level of MMP-9,NF-κB p65 and p50 protein in villi was higher than that in the control group,and the level of IκBβ protein in villi was lower than that in the control group(P<0.05).Conclusion:The increase of TNF-α level in peripheral cells can affect the degradation of the Protein of IκB in villi.The elevated expression of NF-κB increases the level of MMP-9,the excessive degradation of extracellular matrix in decidual leads to the shedding of embryo,and may occur miscarriage eventually. |
