Objective To research the expression of adiponectin receptor T-cadherin in ISO-induced myocardial infarction model in rats and its correlation with acute inflammatory response,so as to explore its early warning effect on acute myocardial infarction.Methods Selected Wistar male rats(SPF level)with body weight of 220-240 g were randomly divided into the Myocardial Infarction model group and the control group according to the randomized number table.Twelve rats in each group were injected with isoprenaline(ISO)150 mg kg-1.d-1 subcutaneously at intervals of 24 hours for 2 consecutive days after recording normal electrocardiograms.The control group was subcutaneously injected with the same amount of saline(0.2ml/100g)for 2consecutive days.After the second subcutaneous injection of isoproterenol for 24 hours,the synchronous six-lead(limb lead)electrocardiogram was monitored to measure the heart rate.Observe Q spread of ST segment deviation degree,lead to Ⅱst-elevation is greater than 0.2 mV or Q waveform became the standard myocardial infarction.After the AMI model was successfully anesthetized in rats,the heart and serum of the rats were taken,the heart tissue was stained with HE to observe the morphology,and the protein expression of t-cadherin(t-cad)in the myocardium was detected by immunohistochemistry.RT-PCR was used to detect T-cadherin gene expression in myocardium.ELISA was used to detect serum adiponectin(ADP,Adiponectin,Acrp30),T-cadherin(T-cad),and hypersensitive C-reactive protein.The contents of hs-CRP and Tumor Necrosis Factor Alpha(TNF-alpha)protein.ResultsThe results of HE staining showed that compared with the control group,the myocardial cell capsule was incomplete in the myocardial infarction group,and the myocardial fibers were thickened,lengthened,disorganized,and interstitial Spaces were widened,the number of nuclei was reduced,and the nuclei were large anddeeply stained,indicating the infiltration of inflammatory cells,indicating the necrosis of myocardial cells.Immunohistochemical results showed that the expression level of t-cadherin in myocardial tissue of the myocardial infarction group was decreased compared with the control group(0.1567±0.0061vs0.1228±0.0027),and the difference between the two groups was statistically significant(P<0.05).RT-PCR results showed that,compared with the control group,the mRNA expression level of t-cadherin in myocardial tissues of the myocardial infarction group was decreased(2.133 ± 0.2385vs1.359 ± 0.1101),and the difference between the two groups was statistically significant(P<0.05).The results of ELISA showed that the serum adiponectin(Acrp30)(ng/mL)levels in the myocardial infarction group were significantly lower than those in the control group(7.016±0.7236 vs1.883±0.2240,P<0.01),and the serum T-cadherin(T-cad)(ng/mL)levels in the myocardial infarctiongroup were significantly higher than those in the control group(6.075 ± 0.8149vs14.61±1.583,P< 0.01).The content of Tumor Necrosis Factor Alpha(TNF-Alpha)(pg/mL)in the myocardial infarction group was significantly increased,and the difference between the two groups was statistically significant(1860±95.96vs3141±92.5,p<0.01).The content of hs-CRP(ng/mL)was significantly higher than that of the control group(20.93±1.079vs30.49±1.742,p<0.01).Serum T-cadherin was negatively correlated with serum adiponectin,serum T-cadherin was positively correlated with inflammatory factors(TNF-alpha,hs-CRP),serum adiponectin was negatively correlated with inflammatory factors(TNF-alpha,hs-CRP),and there was no correlation between T-cadherin and inflammatory factors in rat myocardium.ConclusionISO-induced success of AMI model.The mRNA and protein expression levels of t-cadherin(t-cadherin,CDH13)in the ventricle of the myocardial infarction model group were decreased,while the serum levels of t-cadherin in the myocardial infarction model group were significantly increased.Research on T-cadherin have shown that it may be helpful as a diagnostic marker in the early stage of cardiovascular disease,and may be an important molecular marker of acute myocardial injury,which plays an important role in early warning of disease.Serum adiponectin expression decreased in the early stage of myocardial infarction,whileserum T-cadherin expression increased in the early stage of myocardial infarction.Serum adiponectin was negatively correlated with serum T-cadherin.Serum T-cadherin was positively correlated with inflammatory factors(TNF-alpha,hs-CRP),and serum adiponectin was negatively correlated with inflammatory factors(TNF-alpha,hs-CRP).This experimental study shows that adiponectin has a good anti-inflammatory effect.T-cadherin may participate in the inflammatory response of acute myocardial infarction. |
