Objective:To investigate the effect of chronic stress on the level of PGC-1α/FNDC5/BDNF pathway in mouse muscular brain and the effect of aerobic exercise on the pathway level of muscular brain on the basis of constructing the model of chronic stress and depression mice.Methods:45 male KM mice were randomly divided into blank control group(CG group),model control group(MG group),model motion group(ME group),15 in each group.The MG group and ME group used 12 kinds of chronic stress stimulation for 4 weeks to establish the Depression model.After the model was successfully established,the ME group used the running Table movement mode to carry on the aerobic exercise intervention,where in the slope is 0,the speed 10m/min×60min(the load strength is equivalent to the 60%(1?o2max),the weekly training 6 days,a total of 6 weeks.At the end of the training,neurological behavior assessment was carried out in various mice,and the morphological structure of brain tissue was observed by Nissl staining,and the expression of PGC-1α,FNDC5,BDNF protein and mRNA in brain tissue and quadriceps was detected by Western blot and RT-PCR.Results:1)The results of neurobehavioral evaluation showed that the consumption of sugar water in MG group and the score of horizontal and vertical activity in open box experiment decreased obviously(p<0.01)compared with CG group,and the time of forced swimming and suspension experiment increased obviously(p<0.01).After aerobic exercise intervention,the results of all the indexes of ME group were improved obviously(p<0.01).2)The results of the brain tissue Nissl staining showed that the small body of the MG group was dispersed and irregular,and there were cavitation and nuclear fixation,and the degree of neuronal damage was greater.Compared with MG group,the neurons in the ME group were more concentrated,the cavitation and nuclear fixation were lighter,and the damage of neurons in brain tissue improved.3)The results of brain tissue PGC-1α,FNDC5,BDNF protein and mRNA test showed that the expression of PGC-1α,FNDC5,BDNF protein and mRNA in MG group was significantly reduced compared with CG group(p<0.01),and compared with MG group,There was a significant increase in the expression of factor protein and mRNA in the brain tissue of ME group(p<0.01,p<0.05).4)The results of skeletal muscle PGC-1α,FNDC5 protein and mRNA test showed that the expression of skeletal muscle PGC-1α,FNDC5 protein and mRNA in MG group decreased significantly compared with CG group(p<0.01),and the expression of factor protein and mRNA in skeletal muscle of ME group increased significantly in the comparison MG group(p<0.01);5)The correlation between brain tissue and skeletal muscle PGC-1αand FNDC5 mRNA showed that there was a high positive correlation(r>0.75)between brain tissue and the expression of PGC-1αand FNDC5mRNA in skeletal muscle,which was statistically significant(p<0.01).Conclusion:1)Aerobic exercise can improve depression in mice with chronic stress depression and promote the repair of brain injury.2)It is suggested that moderate intensity aerobic exercise may play an antidepressant role by raising the expression of the target gene BDNF through the muscle and brain PGC-1α/FNDC5 pathway.
