Objective: To systematically evaluate the efficacy of aerobic exercise intervention in depression patients and animal models.Through text mining and bioinformatics analysis,the effective targets,key targets,signaling pathways and mi RNA-m RNA regulatory networks of aerobic exercise intervention in depression were screened to predict the potential mechanism of aerobic exercise intervention in depression.Methods: 1.CNKI,Wanfang database,VIP database,CBM,Pub Med,Cochrane Library,Embase,Web of Science database were searched.The retrieval time was from the establishment of each database to October 2022.The clinical randomized controlled trials and animal controlled trials of aerobic exercise intervention in depression that met the inclusion criteria were screened,and the quality of the literature was evaluated.The statistical software Review Manage 5.3 and R software were used for statistical analysis of the collected data.2.CNKI,Wanfang database,VIP,CBM,Pub Med,Cochrane Library,Embase,Web of Science database were searched for clinical trials and animal experiments of aerobic exercise intervention in depression from the establishment of each database to October 2022.The effective targets of aerobic exercise intervention in depression were extracted from the studies that met the inclusion criteria.The Uniprot and NCBI Gene databases were used to standardize the naming of the collected targets,and the disease targets of depression collected by Gene Cards,Dis Ge NET and NCBI Gene databases were intersected to construct an effective target data set of aerobic exercise intervention in depression.Metascape database was used to analyze gene ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment of effective targets related to aerobic exercise and three exercise modes(treadmill,swimming,and voluntary wheeling).Through STRING database and Cytoscape 3.8.2 software,the protein-protein interaction network of effective targets and the intersection of three exercise-related targets were constructed to obtain key targets,predict mi RNAs upstream of key targets,and construct a potential regulatory network for aerobic exercise intervention in depression.Result: 1.A total of 17 articles and 1111 patients were included in the systematic review of clinical trials.The results of Meta-analysis showed that the scores of Hamilton Depression Scale,Self-rating Depression Scale and Beck Depression Scale in the aerobic exercise intervention group were significantly lower than those in the control group(P < 0.05),the intervention cycle may be the source of heterogeneity between the Hamilton Depression Scale groups..2.A total of 30 articles and 801 samples were included in the systematic evaluation of animal experiments.The results of Meta-analysis showed that compared with the control group,aerobic exercise could reduce the forced swimming latency and tail suspension latency,and increase the sugar preference index,the differences were statistically significant(P < 0.05).Different animal models and modeling methods may be the source of heterogeneity between tail suspension latency groups.The sugar preference index was used as the outcome index to conduct a network meta-analysis of different aerobic exercise interventions for depression.The results showed that the intervention effect of voluntary wheeling was better than swimming and treadmill exercise.3.A total of 90 articles were included in text mining and bioinformatics analysis,and 135 effective targets of aerobic exercise intervention for depression were obtained.The disease database was combined to collect depression-related targets for screening to obtain 110 effective targets,and the data set was constructed according to the movement mode and expression type.Enrichment analysis of 110 effective targets and three aerobic exercise-related effective targets was mainly enriched in inflammatory response,oxidative stress,immune response and apoptosis-related biological functions and PI3K/AKT,MAPK,HIF-1 and IL-17 signaling pathways.Among them,PI3K/AKT signaling pathway is an important signaling pathway for the three aerobic exercises.By constructing a PPI network of effective targets and the intersection of three aerobic exercise-related targets,17 key targets were obtained,namely BDNF,IL6,TNF,CREB1,IL1 B,IL10,IDO1,VGF,GFAP,MAPK1,PPARGC1 A,AKT1,CASP3,VEGFA,STAT3,CTNNB1,JUN.Through mi RWalk,mi RNet and mir DIP databases,297 mi RNAs upstream of17 key targets were predicted,and the mi RNA-m RNA regulatory network of key targets was constructed.The potential regulatory network of aerobic exercise intervention in depression based on text mining and bioinformatics analysis was integrated.Conclusion: 1.Aerobic exercise intervention can reduce the scores of Hamilton depression scale,self-rating depression scale and Beck depression scale,and improve the depressive symptoms of patients.2.Aerobic exercise can improve depression-like behavior in depressed animal models,significantly reduce the forced swimming latency and tail suspension latency,and increase the index of sucrose preference test.The effect of voluntary wheeling on the index of sucrose preference test of depressed animal models was better than swimming and treadmill exercise.3.A dataset of 110 effective targets for aerobic exercise intervention in depression was constructed.Aerobic exercise mainly regulates PI3 K/AKT,MAPK,HIF-1 and IL-17 signaling pathways through key targets such as BDNF,IL6,TNF,CREB1,IL1 B,IL10,IDO1,VGF,GFAP,MAPK1,PPARGC1 A,AKT1,CASP3,VEGFA,STAT3,CTNNB1,JUN,etc.,and participates in biological processes such as inflammatory response,oxidative stress,immune response and apoptosis to exert antidepressant effects.The PI3K/AKT signaling pathway is a key signaling pathway.297 mi RNAs upstream of 17 key targets are predicted,and the mi RNAm RNA interaction network of key targets is constructed.Based on text mining and bioinformatics analysis,the potential regulatory network of aerobic exercise intervention in depression is constructed to provide reference and direction for further researchers. |
