Objectives To observe the expression and distribution of CXCL13,CXCR5 and CXCR5~+CD8~+T lymphocytes in cervical cancer tissues,and to study its clinical significance in cervical cancer.Methods In this study,176 women with different pathological types of cervical tissue were collected from female patients who were treated in the gynecology department of Tangshan Workers’Hospital from September 2014 to September 2017.47 specimens of patients with cervical squamous cell carcinoma(CSCC)were assigned to the CSCC group,45 patients with benign uterine disease and hysterectomy were selected as control group,38 patients with low-grade cervical intraepithelial neoplasia(CIN I)were enrolled in the CINI group,and 46 patients with high-grade cervical intraepithelial neoplasia(CIN II-III)were classified as CIN II-III group.Immunohistochemistry(SP),real-time quantitative RT-qPCR and Western blot were used to detect the expression of CXCL13 and CXCR5mRNA and protein in different pathological cervical tissues and cervical cancer cell lines,and the cervical tissues were analyzed by immunofluorescence co-localization.The distribution of CD8~+T lymphocytes and CXCR5~+CD8~+T lymphocytes was analyzed,and the correlation between CXCL13 mRNA expression and CXCR5~+CD8~+T lymphocyte infiltration and clinical data of CSCC patients was analyzed.Results 1 Statistics of basic clinical information of each group(age,age of menarche,menstrual cycle,menstrual days,number of pregnancies,number of abortions,smoking and drinking)showed no statistical difference between the groups(P>0.05).2 The expression of CXCL13 and CXCR5 mRNA and its protein increased first and then decreased gradually.The expression of CXCL13 and CXCR5 mRNA was the highest in CIN I group,lower in CIN II-III group,and significantly decreased in CSCC group,the difference was statistically significant(P<0.05);Western blot method further confirmed that the protein expression of CXCL13 and CXCR5 was significantly decreased in cervical cancer cell lines(P<0.05).3 Immunofluorescence co-localization revealed that the infiltration of CD8~+T lymphocytes and CXCR5~+CD8~+T lymphocytes increased first and then decreased with the progression of cervical lesions,among which the highest infiltration in CIN I group was in CIN II-III group.The decrease was more significant in the CSCC group,and there were statistical differences among groups(P<0.05).4 The expression level of CXCL13 mRNA and the degree of CXCR5~+CD8~+T lymphocyte infiltration in the CSCC group were not related to the age and tumor size of the patients(P<0.05);and the pathological differentiation of the tumor,lymph node metastasis,tissue invasion degree and clinical stage(P<0.05).Conclusions 1 The expression and infiltration of CXCL13,CXCR5 and CXCR5~+CD8~+T lymphocytes may be involved in the inhibition of the occurrence and development of cervical cancer.2 The expression level of CXCL13 and the degree of CXCR5~+CD8~+T lymphocyte infiltration were related to the size of the tumor,clinical stage,depth of tissue invasion and lymph node metastasis.CXCL13/CXCR5 biological axis and CXCR5~+CD8~+T lymphocytes are expected to be important monitoring indicators for clinical evaluation of cervical cancer patients.Figure5;Table7;Reference 138...
