| Objective:By observing the expression of chemotactic factor CXCL13 and its receptor CXCR5 in the squamous cell carcinoma of larynx tissue and para-carcinoma tissue,we analysed their clinical significance combined with prognosis and the clinical pathological parameters(including gender,age,primary site,the degree of differentiation,TNM stage,clinical stage,lymph node metastasis or not),and explored its clinical significance.Materials and Methods:Collecting the tissue specimens of paraffin embedding that underwent the surgical removal of laryngeal squamous cell carcinoma in the first affiliated hospital of Shanxi medical university from February 2000 to June 2015,we selected 132 cases of laryngeal squamous cell carcinoma paraffin tissue specimens,and the corresponding specimens of 66 cases of normal mucosa of paraffin tissue adjacent to carcinoma(normal mucosa tissue adjacent to carcinoma,5 mm away from the tumor edge).The pathological data of the selected specimens was complete.After the pathological diagnosis,the specimens were all laryngeal squamous cell carcinoma and they primary.Radiotherapy and chemotherapy was not accepted preoperative.Immunohistochemistry(IHC)was used to detect the expression of CXCL13 and CXCR5 protein in the squamous cell carcinoma of larynx and normal mucosa paraffin embedding tissue adjacent to carcinoma.We scanned the completed slices that dealt with immunohistochemical method by digital pathological section scanner.Reading and auditting them with CaseViewer1.4 image acquisition analysis software.The brown and tawny area can be determined as positive expression.SPSS Version20.0 statistical softwarewas used for experimental results analysis.The data was analyzed by χ2 test,Spearman rank correlation analysis,Kaplan Meier-survival analysis and Cox-Regression analysis.Take α=0.05 as examination standard,and P≤0.05 shows statistical significance.Results:1、 CXCL13 and CXCR5 expression in laryngeal squamous cell carcinoma tissues and normal tissues:The positive rate of CXCL13 expression in laryngeal squamous cell carcinoma tissues was 50.8%,3.0% in tissues adjacent to carcinoma.The positive rate of CXCR5 expression in laryngeal squamous cell carcinoma tissues was 45.5%,4.5% in the tissues adjacent to carcinoma.The positive expression of CXCL13 and CXCR5 in laryngeal squamous cell carcinoma tissues compared with normal mucosa tissues adjacent to carcinoma was different,it shows statistical significance(P<0.05).2 、 The correlation between CXCL13 and CXCR5 protein expression and clinical pathological parameters:CXCL13 and CXCR5 protein expression strength was not obviously correlated with gender,age(P > 0.05),which was significantly correlated with TNM stage,lymph node metastasis and clinical stage(P < 0.05);the expression in the glottis carcinoma and supraglottic carcinoma had significant difference(P < 0.05);CXCL13 expression was associated with the degree of differentiation(P = 0.05).3、The relationship between CXCL13 and CXCR5 expression in laryngeal squamous cell carcinoma tissues and correlation with survival time:The expression between CXCL13 and CXCR5 in laryngeal squamous cell carcinoma tissues had significant correlation.The survival time of low CXCL13 and CXCR5 expression patients compared with high expression ones had statistical significance(P <0.05).Conclusions:The positive expression rate of CXCL13 and CXCR5 in laryngeal squamous cell carcinoma tissues was significantly higher than tissues adjacent to carcinoma;we analysed the correlation between clinical pathological parameters with expression level of CXCL13 and CXCR5 and finded that: the expression level of CXCL13 and its receptor protein CXCR5 can be used as a new indicator to evaluate the possibility of invasion and metastasis of laryngeal cancer incidence;by comparing the patients survival time of different expression levels of laryngeal squamous cell carcinoma,reminds that CXCL13 and CXCR5 may be adverse prognostic factor for patients with laryngeal cancer. |
