Objective To investigate the expression of P63,PTEN and Ki67 in the PA tissues and malignant tumor tissues of parotid gland.Analysis the positive expression trend of P63,PTEN and Ki67 in the normal tissues,PA tissues and malignant tumor tissues of parotid gland.To investigate the correlation between the expression of P63,PTEN,Ki67 and Clinical Staging of malignant tumor tissues of parotid gland,combined with the patients’medical records.To analysis the correlation of expression of P63,PTEN and Ki67 in the malignant tumor tissues of parotid gland.Joint detection of the expression of P63,PTEN and Ki67 provides theoretic foundation for evaluating progression of tumors of parotid gland.Methods 1 Materials:We collected paraffin-embedded tissue samples diagnosed by pathology department,in the Hospital of North China University of Science and Technology,from January,2013 to January,2018.The experimental group contained 60paraffin-embeded samples of Pleomorphic adenoma tissues of parotid gland and 60paraffin-embeded samples of malignant tumor tissues of parotid gland.The control group contained 60 normal tissues of parotid gland.None of these patients received treatment,radiotherapy,chemotherapy or hormonotherapy for 3 months before surgery.None of these patients had tumors in other organs of the body.Malignant tumor tissues of parotid gland were classified by the eighth edition of Cancer Staging Manual revised by the AJCC in 2017.2 The expression of P63,PTEN and Ki67 in normal tissues,Pleomorphic adenoma and malignant tumor tissues of the parotid gland was detected by immunohistochemistry.3 SPSS 17.0 statistical software was used for data analysis,and the counting data was tested byχ~2 test or Fisher’s exact test and Spearman rank correlation analysis.P<0.05 showed that difference had statistically significant difference.Results 1 The positive expression rate of P63 in normal tissues,PA tissues and malignant tumor tissues of parotid gland respectively were 30.0%,36.7%and 63.3%.The positive expression rate of PTEN in normal tissues,PA tissues and malignant tumor tissues of parotid gland respectively were 90.0%,35.0%and 26.7%.The positive expression rate of Ki67 in normal tissues,PA tissues and malignant tumor tissues of parotid gland respectively were 41.7%,55.0%and 68.3%.The positive expression rate of P63 and Ki67showed an increasing trend in the normal tissues,PA tissues and malignant tumor tissues of parotid gland.The difference had statistically significant(P<0.05).The positive expression rate of PTEN showed a decreasing trend in the normal tissues,PA tissues and malignant tumor tissues of parotid gland.The difference had statistically significant(P<0.05).2(1)The positive expression rates of P63 in the the two age groups of the malignant tumor tissues of parotid gland were 66.7%and 60.0%.The difference had no statistically significant(P>0.05).The positive expression rates of P63 in the male and female groups of malignant tumor tissues of parotid gland were 54.2%and 69.4%.The difference had no statistically significant(P>0.05).The positive expression rates of P63 in the four T staging of the malignant tumor tissues of parotid gland were 83.3%,83.3%,50.0%and 59.1%.The difference had no statistically significant(P>0.05).The positive expression rates of P63 in three N staging of the malignant tumor tissues of parotid gland were 54.5%,77.8%and 100%.The difference had statistically significant(P<0.05).The positive expression rates of P63 in two M staging of the malignant tumor tissues of parotid gland were 58.3%and 83.3%.The difference had statistically significant(P<0.05).(2)The positive expression rates of PTEN in the two age groups of the malignant tumor tissues of parotid gland were 33.3%and 20.0%.The difference had no statistically significant(P>0.05).The positive expression rates of PTEN in the male and female groups of malignant tumor tissues of parotid gland were 20.8%and 30.6%.The difference had no statistically significant(P>0.05).The positive expression rates of PTEN in the four T staging of the malignant tumor tissues of parotid gland were 83.3%,50.0%,15.0%and 9.1%.The difference had statistically significant(P<0.05).The positive expression rates of PTEN in three N staging of the malignant tumor tissues of parotid gland were 29.5%,22.2%and14.3%.The difference had no statistically significant(P>0.05).The positive expression rates of PTEN in two M staging of the malignant tumor tissues of parotid gland were33.3%and 0.The difference had statistically significant(P<0.05).(3)The positive expression rates of Ki67 in the the two age groups of the malignant tumor tissues of parotid gland were 73.3%and 63.3%.The difference had no statistically significant(P>0.05).The positive expression rates of Ki67 in the male and female groups of the malignant tumor tissues of parotid gland were 58.3%and 75.0%.The difference had no statistically significant(P>0.05).The positive expression rates of Ki67 in the four T staging of the malignant tumor tissues of parotid gland were 16.7%,58.3%,60.0%and 95.5%.The difference had statistically significant(P<0.05).The positive expression rates of Ki67 in three N staging of the malignant tumor tissues of parotid gland were 79.5%,22.2%and57.1%.The difference had statistically significant(P<0.05).The positive expression rates of Ki67 in two M staging of the malignant tumor tissues of parotid gland were 60.4%and100%.The difference had statistically significant(P<0.05).3 The positive expression rate of P63 and PTEN in the malignant tumor tissues of parotid gland was negatively correlated with the correlation coefficient of-0.245.The correlation had statistically significant(P<0.05).The positive expression rate of Ki67 and PTEN in the malignant tumor tissues of parotid gland was negatively correlated with the correlation coefficient of-0.400.The correlation had statistically significant(P<0.05).The positive expression rate of P63 and Ki67 in the malignant tumor tissues of parotid gland was positively correlated with the correlation coefficient of 0.746.The correlation had statistically significant(P<0.05).Conclusions 1 The positive expression rate of P63 and Ki67 showed an increasing trend in the normal tissues,PA tissues and malignant tumor tissues of parotid gland.The positive expression rate of PTEN showed a decreasing trend in the normal tissues,PA tissues and malignant tumor tissues of parotid gland.2 The positive expression rates of P63 were related to lymphatic metastasis and distant metastasis.The higher the N and M stages were,the higher the positive expression rate of P63 was.The positive expression rates of PTEN were related to tumor size and distant metastasis.The higher the T and M stages were,the lower the positive expression rate of PTEN was.The positive expression rates of Ki67 were related to tumor size,lymphatic metastasis and distant metastasis.The higher the T,N and M stages were,the higher the positive expression rate of Ki67 was.3The positive expression rate of P63 and PTEN in the malignant tumor tissues of parotid gland was negatively correlated.The positive expression rate of Ki67 and PTEN in the malignant tumor tissues of parotid gland was negatively correlated.The positive expression rate of P63
