Objectives By establishing a rat model of dry socket osteoporosis,strontium ranelate was used to intervene in the healing process of dry socket osteoporosis.The changes of bone trabeculae and the expressions of bone morphogenetic protein-2(BMP-2)and tartrate-resistant acid phosphatase(TRAP)were observed at different stages of tooth extraction.To explore whether strontium ranelate can accelerate the healing of extraction socket in osteoporotic patients after suffering from dry socket.To provide experimental basis for the treatment of osteoporosis complicated with dry socket combined with systemic medication.Methods Fifty-four female SD rats were selected and weighed(240±20)g at the age of three months.Rats were randomly divided into blank control group(group A),osteoporosis group(group B),strontium ranelate group(group C),18 rats in each group.Osteoporosis model was established in rats of osteoporosis group(group B)and strontium ranelate group(group C).Rats in osteoporosis group(group B)and strontium ranilate group(group C)were given 80mg/(kg·d)of retinoic acid by intragastric administration for 15 days,while rats in blank control group(group A)were given the same amount of distilled water by intragastric administration for 15 days.Six rats in each group were randomly selected after gastric administration.Bone mass density(BMD)of femur was measured by dual-energy X-ray to test whether the model of osteoporosis was successfully established.After successful modeling,all three groups of rats were extracted unilateral maxillary first molars at the same time.Thirty minutes after extraction,the blood clot in the extraction socket was removed and the hemostasis was stopped by wiping the extraction socket with adrenaline cotton ball immersed in 1:1000concentration.Then the cotton ball with rat saliva was applied to the extraction socket to infect the extraction socket.Three days later,we observed whether the rat model of dry socket syndrome was successfully established.After dry socket in rats,rats in the strontium ranelate group(group C)were given intragastric administration of strontium ranelate at a dose of 400 mg/(kg·d)to treat osteoporosis,while rats in the blank control group(group A)and osteoporosis group(group B)were given intragastric administration of distilled water to control the experiment.Six rats in each group were randomly executed on the 7th,14 th and 28 th day after intragastric administration.Histological changes of alveolar bone were observed by hematoxylin-eosin staining.Immunohistochemical method was used to detect the expression of BMP-2 and TRAP staining was used to detect the expression of osteoclasts.Image-Pro-plus 6.0 image analysis system software was used to semi-quantitatively analyze the expression level of BMP-2 and TRAP staining.The measurement data of the two factors were processed by SPSS22.0 software.The results were expressed in the form of mean±standard deviation(x ±s),and the comparison between groups was performed by one-way ANOVA.There was a significant difference in the mean optical density of BMP-2 and TRAP staining between the two groups,when P<0.05.Results Osteoporotic rats were killed at 7,14 and 28 days after suffering from dry socket.Morphological observation showed that the bone trabeculae of extraction socket increased and the healing of extraction socket was better in the blank control group than in the osteoporosis group at the same time.The bone trabeculae in strontium ranilate group were more than those in osteoporosis group.At the same time,BMP-2 expressionin blank control group and strontium ranelate group was higher than that in osteoporosis group,and the number of osteoclasts with tartrate-resistant acid phosphatase positive expression was lower than that in osteoporosis group.The difference was statistically significant(P<0.05).Conclusions 1 The experiment proved that osteoporosis could delay the bone healing of extraction socket in dry socket.2 Strontium ranelate can increase the expression of BMP-2and decrease the expression of TRAP during the healing of dry socket in osteoporotic rats.Strontium ranelate can promote the healing of extraction sockets of dry sockets in osteoporotic rats.Figure33;Table6;Reference 131...
