Effects Of Orthodontic Tooth Movement On Strontium Ranelate And Qianggu Capsule For Osteoporosis Rats

Objective: By establishing the experimental animal model of osteoporosis rats with medical intervention to investigate the change of orthodontic tooth movement distance in osteoporosis rats after application of stress; to observe changes on contents of Calcium(Ca), Phosphorous(P) and Alkaline Phosphatase(ALP) in blood; to test the effect on the tooth movement experimental process of osteoporotic changes in IL-1β. Providing experimental basis for clinical treatment of osteoporosis patients via measuring the tooth movement speed、evaluating the risk of root resorption and medical efficacy with the experimental results.Method: Place sixty, four-week-old, weight of(210±10)g SD rats(male) were freely in the SPF breeding environment. The rats were assigned into four groups in total(n=15 in each) randomly and equally, which were named control group, osteoporosis group, Qianggu capsule group and strontium ranelate group. Rats in the last three groups mentioned above were induced by retinoic acid in 80mg/(kg·d) for two weeks, rats in control group were administered distillated water, also for two weeks. Followed that, detected the rat tibial bone mineral density and confirm the success of modeling. After successful modeling, rats in Qianggu group were induced by Qianggu capsule solvent in 90mg/(kg·d); rats in strontium ranelate group were induced by strontium ranelate solvent in 400mg/(kg · d); rats in control group and osteoporosis group were administered distillated water as the controlled experiment. The Orthodontic tooth movement devices were installed for all rats after two weeks of medical intervention treatment. Five rats were sacrificed at the end of 7, 14, 21 days after application of stress, randomly and respectively. Measure the first molar tooth movement distance for rats with forcing, detect contents of Ca, P and Alkaline ALP in blood, observe morphological changes of rat periodontium and tooth root through Hematoxylin-Eosin staining(H-E staining). Method of Immunohistochemical staining was employed for the first molar periodontium of rats with forcing received. The semi-quantitative analysis for the expression of IL-1β was made by image analysis system. Based on the histological observation and image analysis, analyzed statistically on the gray values.Result: Regardless of forcing time for orthodontic tooth movement, the tooth movement distance for rats in osteoporosis group was greater than it in control group, the difference was statistically significant(P<0.05); after the medical treatment, compared to rats in osteoporosis group, rats in Qianggu capsule group and strontium ranelate group could not cause the change of tooth movement distance、change of contents of Ca, P and ALP in blood, the difference was not statistically significant(P>0.05), in the meantime, the vary in IL-1β was not obviously(P>0.05) at the end of 7 days after application of stress; at the end of 14 days, the tooth movement distance reduced(P<0.05), contents of Ca, P and ALP in blood was higher(P<0.05) but IL-1β decreased obviously(P<0.05)for rats in strontium ranelate group compared with rats in osteoporosis group; all the results above changed for rats in Qianggu capsule group, but the difference was not statistically significant(P>0.05); at the end of 21 days, the tooth movement distance reduced, contents of Ca, P and ALP in blood increased and the IL-1β in Periodontium decreased for rats in Qianggu capsule group, strontium ranelate group compared with rats in osteoporosis group, and the difference was statistically significant(P<0.05).Conclusion: 1. The osteoporosis rats have a faster tooth movement speed, a higher IL-1β in Periodontium and a higher risk of root resorption for side of forcing tooth than those of normal rats. 2. Qianggu capsule and strontium ranelate both have positive effects on osteoporosis rats, such as reducing the tooth movement speed, increasing contents of Ca, P and ALP in blood, decreasing the level of IL-1β in Periodontium and lowering risk of root resorption. 3. The appearance of treatment efficacy for medicine strontium ranelate is proceeding to that of Qianggu capsule.
