Objective:To research whether the petroleum ether extracts of Anthriscus sylvestris(PEAS)could inhibit inflammatory environment in colon cancer by inhibiting signaling pathway of STAT3;To discuss the mechanism of PEAS improve inflammatory environment of colon cancer mice;To discuss the mechanism of PEAS affect inflammatory HT29 cell.Method:(1)to tese the inhibition effect of PEAS for HT29 cell and the relation between effect and concentration or time.(2)to establish the model of ulcerative colitis-related colorectal cancer(UCRCC),analyse the effect of PEAS to UCRCC mice by grade,weight,the thymus index and the spleen index.(3)to tese expression of related protein of HT29 cell and colon tissue by Western Blot.(4)to analyse the colon tissue by immunohistochemical.(5)to tese expression of IL-1β,IL-6,TNF-αafter using PEAS by ELISA.Results:(1)PEAS could control the development of HT29 cell,and the effect will improve when increaseconcentration,IC50=25.180μg/ml.(2)PEAS could control the expression of STAT3 relatedprotein,like p-STAT3,NF-κB,JAK1,JAK2,SHP-1,SHP-2,and the effect is related with concentration and time.(3)PEAS could improve health condition and survival rate,and could inhibit the expression of STAT3,p-STAT3,BCL-2,MCL-1,NF-κB,XIAP.(4)establishing inflammation HT29 cell model,and proving PEAS could inhibit the expression of IL-1β,IL-6,TNF-α.Conclusion:PEAS could inhibit the development of colon cancer by STAT3 signal channel.
