| Objective: To observe the effects of Wenyang Xiezhuo Tongluo Recipe on TGF-β1 and JAK2/STAT3 signaling pathways in renal tissues of 5/6nephrectomized rats,and to explore the possible mechanism of improving renal fibrosis and protecting renal function.Methods:75 healthy male Sprague-Dawley rats were randomly divided into sham operation group(C group)and model group.In the model group,5/6nephrectomy was performed to replicate chronic renal failure in rats.After successful modeling,the model rats were randomly divided into model group(M group)and losartan potassium group(X group),niaoduqing group(Q group)and Chinese medicine group(Z group),each treatment group were given corresponding drugs,and were continuously administered for 12 weeks.Renal pathological changes were observed;renal function was detected;mRNA and protein expressions of TGF-β1 and JAK2/STAT3 signaling pathways in renal tissues were detected by RT-PCR and Western Blot.Statistical analysis of the experimental data was performed by using One-way ANOVA.Results:1 Observation results of kidney morphology under light microscopyThe renal structure of group C was clear and normal;Group M glomerular mesangial cells and stromal hyperplasia,glomerular sclerosis,renal interstitial fibrosis with mononuclear cell infiltration;Compared with M,the damage was improved to varying degrees in each treatment group.Masson showed a reduction in the degree of renal interstitial fibrosis,and the decrease in the group Z was the most significant(P<0.05).2 Comparison of renal function in ratsCompared with group C,BUN,Scr and UA in group M increased(P<0.05);Compared with group M,BUN,Scr and UA in group Q and group Z decreased(P<0.05),of which group Z UA Significantly decreased(P<0.01);Compared with Q group,Z group BUN,Scr and UA decreased(P>0.05).3 Expression of TGF-β1,JAK2 and STAT3 mRNA in rat kidneyCompared with group C,the expression of TGF-β1,JAK2 and STAT3 mRNA in group M was significantly increased(P<0.01).Compared with group M,the expression of each group was decreased(P<0.05);Compared with group Q,Z group JAK2 decreased(P <0.05).4 Expression of TGF-β1,JAK2,STAT3 and p-STAT3 proteins in rat kidneyCompared with group C,the expression of TGF-β1,JAK2,STAT3 and p-STAT3 protein in group M was significantly increased(P<0.01).Compared with group M,the expression of TGF-β1,JAK2,STAT3 and p-STAT3 decreased in all treatment groups(P < 0.05).Compared with group X,TGF-β1decreased in group Z(P<0.05).Compared with group Q,JAK2 in group Z decreased(P<0.05).Conclusions:1.Wenyang Xiezhuo Tongluo Recipe can improve the survival of 5/6nephrectomized rats,protect renal function and reduce renal pathological damage;2.Wenyang Xiezhuo Tongluo Recipe can inhibit the expression of TGF-β1 and reduce the accumulation of ECM by inhibiting JAK2/STAT3 signaling pathway,which may be one of the mechanisms of its delay in renal pathological process;3.In the protection of renal function and improvement of renal fibrosis,Wenyang Xiezhuo Tongluo Recipe is superior to losartan potassium and niaoduqing granules. |
