Objective:To observe the association between the expression of Jak2 and Stat3 in perapical in rats.Methods:Twenty five8-week-old SD rats were devided to two groups randomly:A group was Twenty rats as experimental group and five rats in B group for control.Apical periodontitis was induced in A group by exposing their pulp of their right mandibular first molars and enclosed with little PBS cotton ball,then filling with glass ionomer,and adjusting occulsal relationship at last.After the exposure,the rats were sacrificed on day 7、14、21 and 28.The jaws contained right first molar were stripped and prepared for HE staining,polyclonal antibody Jak2、Stat3、p-Jak2 and p-Stat3 were used for immunohistochemsitry,and anti-tartaric acid phosphatase(TRAP)staining was used for detecting the expression of osteoclast.Results:1.HE staining:There is a few inflammatory cells in the periapical area on day7.From 7 to 14 day,the pulp was necrosed and the inflammatory cells was increased heavily.At the 21 day,the lymphocyte was increased and the periapical bone was start to resorb.On day 28,the Howship lacunac was appeared,and the lymphocyte keep increasing.2.immunohistochemsitry staining: A few Jak2-postive cells and Stat3-postive cellswere observed in normal periapical tissue and less p-Jak2、p-Stat3-postive cells were observed.The postive cells of them were both increased at 7 day(P<0.05).Then they both climaxed on day 14(P<0.05).On the day 21-samples,although the expression of Jak2、Stat3、p-Jak2、p-Stat3 reduced,but the expression of them was still higher than the control group(P<0.05).On day 28 the decrease of Jak2、Stat3、p-Jak2 and p-Stat3 was obvious.JAK2 expressed higher than the normal group(P < 0.05).The other’sexpression has no difference between the control(P>0.05)3.TRAP staining:From Day7 to Day 14,the expression of osteoclast was increasing,the acem is on the Day 14.Fewer osteoclasts were observed in the 21-and28-day samples.Conclusion:These researchs demonstrated that Jak2 and Stat3 could be observed in periapical aera,which may be associated with periapical lesions and the generate of osteoclast.
