Objective: To investigate the effect of vitamin B6 on lipid metabolism in mice with nonalcoholic fatty liver disease(NAFLD)and its possible signaling pathway.Methods: Male B129 mice were randomly divided into normal diet group(n=9),high fat diet group(n=8),high fat + vitamin B6(50mg/kg/d)diet group(n=7).After 8 weeks of feeding,the mice in each group were given glucose tolerance test,insulin tolerance test and body weight measurement.The mice were executed and their white fat,serum and liver samples were taken.The abdominal white fat weight,hepatocyte steatosis and lipid content,hepatic inflammation index,hepatic IκB-αprotein,lipid level,serum inflammatory factor level,serum alanine aminotransferase and glutamic oxaloacetate aminotransferase were measured and analyzed in three groups of mice.Result: 1.Compared with normal diet group,hyperlipidemia group had higher blood glucose peak value after injection,higher blood glucose that 2 hours after injection and lower insulin sensitivity(P < 0.05);hyperlipidemia + vitamin B6 diet group had lower blood glucose peak value after injection,lower blood glucose that 2 hours after injection and higher insulin sensitivity(P < 0.05),but compared to than normal diet group,it has higher glucose peak blood glucose after injection,and decreased insulin sensitivity(P < 0.05).2.Compared with the normal diet group,the body fat rate,white fat,hepatocyte steatosis and lipid deposition of mice in the high fat diet group increased,and the levels of total cholesterol,triglyceride and glutamic oxaloacetic transaminase increased(P < 0.05).In the high fat + vitaminB6 diet group,the weight of body weight,white fat weight,hepatocyte steatosis and lipid deposition decreased,while the levels of total cholesterol,triglyceride and glutamic oxaloacetic transaminase decreased(P < 0.05).3.The levels of serum inflammatory factors TNF-a,IL-6,4-HNE and 3-NT in liver of mice fed with high-fat diet group were higher than those of mice fed with normal diet group,and the inhibitory protein I-kappa B-a related to the inflammatory pathway of NF-kappa B was lower,while the levels of serum inflammatory factors and liver inflammatory factors in mice fed with high-fat + vitamin B6 diet group were lower and I-kappa B-a was higher.Conclusion: 1.Vitamin B6 can improve metabolism of blood sugar and lipid to suppress non-alcoholic fatty liver.2.Vitamin B6 can suppress the immune inflamma-tion response of liver by inhibiting IκK/NF-κ B pathway,and inhibit the occurrence and development of non-alcoholic fatty liver disease.
