| Objective:To investigate the effects of moxibustion at different temperatures on the concentrations of glucose,lactate,pyruvate and glutamate in the local tissue microdialysate of Zusanli acupoint in mice under physiological and pathological conditions,in order to provide clues for the initiation mechanism of warm effect of moxibustion.Methods: Physiological status group: 39 male C57BL/6J mice were randomly divided into blank group,(38 ±1)℃-30 min group,(42 ±1)℃-30 min group and(46±1)℃-30 min group with 6 mice in each group.(38 ±1)℃-72 h group,(42 ±1)℃-72 h group,(46 ±1)℃-72 h group,5 rats in each group.Pathological status group: 37 male C57BL/6J mice were used to establish colitis model with 3% DSS drink.after the successful establishment of colitis model,they were randomly divided into model group.Model-(38 ±1)℃-30 min group,model-(42 ±1)℃-30 min group,model-(46±1)℃-30 min group,6 rats in each group;Model-(38 ±1)℃-72 h group,model-(42±1)℃-72 h group,6 rats in each group,5 rats in model-(46 ±1)℃-72 h group.In addition to the blank group and the model group,the local tissue microdialysate of acupoints was collected at 30 min,and 72 h after moxibustion at 38℃,42℃ and46℃ for 30 min and 72 h,respectively.The collection time was 30 min.The concentrations of glucose,lactate,pyruvate and glutamate in local tissue fluid were measured by microdialysis analyzer in real time.Results:1.Physiological state(1)At 30 min after moxibustion,46℃ moxibustion could significantly increase the concentrations of glucose,pyruvate and glutamate in acupoint site tissue microdialysate,but there was no significant change in 42℃ moxibustion group and38℃ moxibustion group(P<0.05),and compared with 46℃ moxibustion,46℃ moxibustion could significantly increase the concentrations of glucose,pyruvate and glutamate in acupoint site tissue microdialysate,but there was no significant change in 42℃ moxibustion group and 38℃ moxibustion group.While lactate increased significantly at 38℃ and 42℃ moxibustion(P<0.05).(2)At 72 h after moxibustion,46℃ moxibustion could significantly increase the concentrations of glucose,lactate,pyruvate and glutamate in the microdialysate of acupoints in the blank moxibustion group were significantly higher than those in the blank group.The concentrations of lactate,pyruvate and glutamate in 42℃ moxibustion group were significantly higher than those in 38℃ moxibustion group(P<0.05),and the concentrations of lactate,pyruvate and glutamate in 42℃ moxibustion group were significantly higher than those in 38℃ moxibustion group(P<0.01).The concentrations of lactate,pyruvate and glutamate in acupoint site tissue microdialysate 72 hours after moxibustion at 46℃ were significantly higher than those in 30 min after moxibustion.2.Pathological state(1)At 30 min after moxibustion,compared with the model group,the concentrations of glucose,lactate,pyruvate and glutamate in the acupoint site tissue microdialysate in the model moxibustion group were higher than those in the model group at 30 min,after moxibustion(P<0.05).Compared with 38℃ moxibustion,the concentrations of glucose and glutamate in acupoint site tissue microdialysate increased significantly at 42℃ and 46℃ moxibustion(P<0.05).Compared with42℃ moxibustion,the concentrations of glucose and lactate increased significantly under 46℃ moxibustion(P < 0.05).(2)At 72 h after moxibustion,the concentrations of lactate and pyruvate in the local tissue microdialysate of the model moxibustion group were higher than those of the model group(P<0.05).Compared with moxibustion at 38℃ and 42℃,the concentrations of glucose,lactate,pyruvate and glutamate in 46℃ moxibustion were significantly higher than those in 38℃ and 42℃ moxibustion(P<0.01).The concentrations of glucose,pyruvate and glutamate in 42℃ moxibustion were significantly higher than those in 38℃ moxibustion(P<0.05).Compared with 72 h after moxibustion,the concentrations of glucose and lactate in local tissue microdialysate at 72 h after moxibustion in model moxibustion group were significantly lower than those in 30 min(P<0.05).The pyruvate in local tissue microdialysate of acupoints at 72 h after moxibustion at 38℃ was significantly lower than that at 30 min(P<0.05).3.Physiological and pathological states.(1)At 30 min after moxibustion,the effects of moxibustion at 38℃ on glucose,lactate,and pyruvate were similar in both physiological and pathological states.However,under pathologica state,38℃ could significantly increase the contents of glutamate(8 times as much as that under physiological condition),and 42℃ moxibustion could significantly increase the contents of glucose,lactate and glutamate.Moxibustion at 46℃ could also significantly increase the contents of glucose,lactate and glutamate,which were 2.6 times,8.8 times and 11 times of physiological state,respectively.(2)At 72 h after moxibustion,the effects of moxibustion at 38℃ on glucose,lactate and pyruvate were similar in both physiological and pathological states.However,under pathological state,38℃ could significantly increase the contents of glutamate(2.3 times as much as that under physiological condition),and 42℃ moxibustion could significantly increase the contents of glucose,glutamate and pyruvate.The contents of glucose,lactate,glutamate and pyruvate were significantly increased by46℃ moxibustion,which were 1.6,1.3,2 and 1.3 times of those in physiological state,respectively,and the contents of glucose,lactate,glutamate and pyruvate were significantly increased by 46℃ moxibustion,which were 1.1 times,2.6 times and 1.2times of those in physiological state,respectively.Conclusions:1.Under different states,moxibustion at different temperatures could increase the concentrations of glucose,lactate,pyruvate and glutamate in acupoint site tissue microdialysate,and the effects of 46℃ moxibustion on lactate and pyruvate was greater than that of 38℃ and 42℃ moxibustion.2.The contents of glucose,lactate,pyruvate and glutamate in acupoint microdialysate before moxibustion were not significantly different from those in physiological state,but the changes of the above substances were obvious after moxibustion.3.After moxibustion at different temperatures,glucose,lactate,pyruvate and glutamate in acupoint site tissue microdialysate changed rapidly,but with the prolongation of time,the change trend decreased. |
