Indigo Naturalis is a dried powder from the leaves or stems of Baphicacanthus cusia(Nees)Bremek.The content of indigo,indirubin and other active components in traditional Indigo Naturalis is low.The content of indigo and indirubin in commercially sold traditional Indigo Naturalis were about 2.5%and 0.27%respectively.Baphicacanthus cusia(Nees)Bremek witch growing in Fujian was selected as raw material in this paper.According to the processing principle of Indigo Naturalis,we have developed a new processing technology Indigo Naturalis.The new processing technology is as follows:Cut the fresh leaves of Baphicacanthus cusia(Nees)Bremek to 12 cm,extract it in hot water,filter and remove leaf residue,add O3 in the filtrate and the filtrate was hydrolyzed with acid solution,after hydrolysis,add alkali solution to adjust pH value to 68,discard the supernatant by standing still,wash the precipitate to neutral,then dry in the oven,and we get the Indigo Naturalis new products.The content of indigo and indirubin in the Indigo Naturalis new products can be increased by 1020 times and 2030 times respectively,the quality of Indigo Naturalis has a qualitative leap.This paper investigates the influence of factors such as O3 time,hydrochloric acid concentration,extraction temperature and extraction time on the new processing technology of Indigo Naturalis.On the basis of single factor experiment,Box-Behnken response surface method was used to optimize the new processing technology of indigo and indirubin respectively.We can adjust and control the direction of Indigo Naturalis according to the market demand.The new processing conditions for directional generation of indigo:the material ratio is 1:10,the hydrochloric acid concentration is 3.4%,the extraction temperature is 79℃,and the extraction time is 33 min,the highest yield of indigo is 2.07 mg/g.The new processing conditions for directional generation of indirubin:the material ratio is 1:10,the hydrochloric acid concentration is 3%,the O3 time is 73 s,the extraction temperature is 79℃,and the extraction time is 33 min,under these conditions,the highest yield of indirubin is 0.758 mg/g.The characteristic fingerprint of Indigo Naturalis was established based on HPLC.Analysis of chemical composition differences between the traditional Indigo Naturalis and Indigo Naturalis new products by chemometric analysis.Analysis of common components in the traditional Indigo Naturalis and Indigo Naturalis new products based on UPLC-Q-TOF-MS/MS Technology.The quality evaluation system of Indigo Naturalis new products is preliminarily established on the basis of their characters,identification,examination,content determination and characteristic fingerprint.To summarize,we developed a new processing technology for processing Indigo Naturalis,the content of indigo and indirubin in Indigo Naturalis new products has increased effectively.We initially established the quality evaluation system of Indigo Naturalis new products.And it’s preliminarily verified by CCK-8 method that the Indigo Naturalis new products have better antitumor activity than the traditional Indigo Naturalis.It provides a favorable reference for the development and utilization of Indigo Naturalis.
